Self-Assembled Aza-BODIPY and Iron(III) Nanoparticles for Photothermal-Enhanced Chemodynamic Therapy in the NIR-II Window

Summary: In this study, a three-dimensional network structure was constructed using metal coordination between catechol-functionalized aza-boron dipyrromethenes and iron ions. The resulting ABFe nanoparticles (NPs) showed intense absorption in the second near-infrared (NIR-II) range and excellent photothermal stability. Additionally, ABFe NPs were capable of generating reactive oxygen species for chemodynamic therapy, which was enhanced by their photothermal performance. The in vitro and in vivo experiments demonstrated the potential of ABFe NPs in photoacoustic imaging and photothermal-enhanced chemodynamic therapy under NIR-II laser irradiation.

Summary: Using fluorescence-guided surgery (FGS) with NIR-II dyes targeting human epidermal growth factor receptor 2 (HER2)-positive ovarian tumors showed favorable tumor accumulation and selective targeting of HER2 positive tumors. These bioconjugated NIR-II-emitting dyes displayed prolonged stability in serum and maintained affinity toward HER2 in vitro, indicating their potential for NIR-II FGS in the cancer setting. Biomimetic Approach to Promote Cellular Uptake and Enhance Photoacoustic Properties of Tumor-Seeking Dyes

Summary: The attachment of glucose to drugs and imaging agents enables cancer cell targeting via interactions with GLUT1 overexpressed on the cell surface. We have developed a biomimetic multivalent glucose moiety (mvGlu) to overcome the limitations of solubility and spectral unmixing in imaging agents, leading to significant enhancements in photoacoustic (PA) signal. We have also demonstrated the utility of mvGlu in targeting cancer cells with ultra-low dye concentrations and in developing a logic-gated acoustogenic probe for detecting intratumoral copper.

Panchromatic small-molecule organic solar cells based on a pyrrolopyrrole aza-BODIPY with a small energy loss

Summary: The authors present two new pyrrolopyrrole aza-BODIPY (PPAB) based panchromatic chromophores, TT-2PPAB and DFBT-2PPAB, with an acceptor-donor-acceptor configuration. Among them, TT-2PPAB exhibits a moderate power conversion efficiency (PCE) of 3.11% when combined with [6,6]-phenyl-C71 butyric acid methyl ester (PC71BM) acceptor, with an energy loss of 0.48 eV, smaller than the empirical limit of 0.6 eV. The efficient photovoltaic performance of the TT-2PPAB-based device is attributed to the sufficiently deep LUMO level of TT-2PPAB for charge transfer to PC71BM, as revealed by electrochemistry and theoretical calculations.

A supramolecular near-infrared nanophotosensitizer from host-guest complex of lactose-capped pillar[5]arene with aza-BODIPY derivative for tumor eradication

Summary: A novel dual phototherapeutic aza-BODIPY supramolecular vesicle was constructed by forming an amphiphilic complex between lactose-capped pillar[5]arene (LacP5) and an aza-BODIPY derivative (BSTA). The supramolecular vesicles showed good stability under physiological conditions, active targeting to liver cancer cells, stimuli-responsive drug release, and dual-mode imaging in vivo (fluorescence and photothermal). The release of aza-BODIPY and disulfiram (DSF) drugs from the vesicles in response to glutathione (GSH) in tumor cells allowed for dual phototherapy and chemotherapy. In vivo antitumor experiments demonstrated enhanced antitumor effects and good biosafety of the dual phototherapy and chemotherapy combination. The LacP5 superset of BSTA supramolecular phototherapeutic vesicles hold great potential in tumor therapy.

Femtosecond Spectroscopy and Nonlinear Optical Properties of aza-BODIPY Derivatives in Solution

It was found that nonfluorescent derivatives exhibited optical gain in the near infrared range under femtosecond excitation. The study also discussed the role of bromine substituents in the molecular structures of these derivatives, and the excited states involved in electronic transitions were investigated using quantum-chemical TD-DFT calculations. The high two-photon absorption and excited-state absorption cross-sections, along with the measured photostability, suggest potential applications in photonics and optical limiting.

Near-infrared vinyl-containing aza-BODIPY nanoparticles as photosensitizer for phototherapy

This compound was then used to prepare asymmetric vinyl-containing azaBDPs 1-8, avoiding the use of Knoevenagel condensation. Discovery of an Amino Acid-Modified Near-Infrared Aza-BODIPY Photosensitizer as an Immune Initiator for Potent Photodynamic Therapy in Melanoma

Summary: In this study, a series of amino acid-modified aza-BODIPY photosensitizers were synthesized and evaluated for photodynamic therapy. Among them, the aspartic acid-modified PS 1a showed the most potent efficacy with enhanced water solubility, increased O-1(2) generation efficiency, and improved photo-dark toxicity ratio. Subcellular localization, cell-based ROS production, and cell death pathway of 1a were studied, and in vivo assays demonstrated its reasonable distribution and clearance. Compared to other PSs, 1a not only exhibited excellent PDT effect with a low drug dose and single-time photoirradiation, but also induced antitumor immunological response.

Near-Infrared fluorescent unsymmetrical aza-BODIPYs: Synthesis, photophysics and TD-DFT calculations

Summary: In this study, the synthesis and properties of four unsymmetrical aza-BODIPYs with near-infrared (NIR) fluorescence were reported. These molecules exhibited highly desirable photophysical and photochemical properties due to the presence of both strongly electron-withdrawing p-nitrophenyl rings and mildly electron-donating p-methoxyphenyl rings within the aza-BODIPY core. The unsymmetrical aza-BODIPYs showed NIR emission with wavelengths ranging from 699 nm to 718 nm, along with fluorescence quantum yields ranging from 0.22 to 0.49 and fluorescence lifetimes ranging from 1.90 ns to 3.59 ns. A specific aza-BODIPY with electron-donating and electron-withdrawing substituents at the 3 and 5 positions, respectively, was identified as a cell-permeable, NIR emitting fluorophore suitable for bioimaging.

Asymmetric AZA-BODIPY with Optical Gain in the Near-Infrared Region
