High-dimensional and single-cell transcriptome analysis of the tumor microenvironment in angioimmunoblastic T cell lymphoma (AITL)

Joshua C. Pritchett et al.

Summary: AITL is an aggressive lymphoid malignancy with a unique tumor microenvironment characterized by a minority population of malignant TFH cells and a diverse infiltrate of immune cells. Non-malignant populations within the TME, such as B cells and CD8+ T cells, are increasingly recognized for their potential role in AITL biology. These non-malignant populations exhibit specific expression patterns that may contribute to the understanding of AITL pathogenesis.

A single-cell atlas of non-haematopoietic cells in human lymph nodes and lymphoma reveals a landscape of stromal remodelling

Yoshiaki Abe et al.

Summary: This study constructed a single-cell transcriptome atlas of non-haematopoietic cells (NHCs), including mesenchymal stromal cells and endothelial cells, collected from human samples. The atlas revealed 30 distinct subclusters of NHCs, including previously unrecognized ones. Comparative analyses with lymphoma NHCs showed subcluster-specific changes in gene expression and interaction with malignant cells in follicular lymphoma NHCs, providing insights into stromal remodelling in lymphoma and potential clinical biomarkers. The study updates NHC taxonomy in human lymph nodes and provides a rich resource for advancing lymphoma management and therapy.

Clonal germinal center B cells function as a niche for T-cell lymphoma

Manabu Fujisawa et al.

Summary: This study investigates the mechanism by which TET2-mutant immune cells enable the development of Angioimmunoblastic T-cell lymphoma (AITL). The results reveal that Tet2-deficient immune cells play a crucial role in AITL development, and aberrant GCB cells in AITL undergo clonal evolution with recurrent mutations in genes related to core histones. Cd40-Cd40lg is identified as a possible mediator of interactions between GCB and tumor cell clusters.

A pan-cancer single-cell transcriptional atlas of tumor infiltrating myeloid cells

Sijin Cheng et al.

Summary: This study conducted a pan-cancer analysis of single myeloid cells from 210 patients across 15 human cancer types, revealing distinct features of TIMs in different cancer types. For example, mast cells in nasopharyngeal cancer were associated with better prognosis, and pro-angiogenic TAMs showed diverse markers across different cancer types. The composition of TIMs appeared to be linked with certain features of somatic mutations and gene expressions.
