Exploring paclitaxel-loaded adenosine-conjugated PEGylated PLGA nanoparticles for targeting triple-negative breast cancer

Summary: In this study, PTX-loaded adenosine-conjugated PLGA nanoparticles were developed for treating TNBC. The nanoparticles showed a small size, high entrapment efficiency, and release at a lower pH. They also exhibited lower hemolysis and improved cellular uptake and anti-TNBC activity compared to free PTX. The nanoparticles had a favorable pharmacokinetic profile and reduced tumor burden without causing significant changes in kidney and liver function biomarkers or histological alterations. Overall, the optimized nanoparticles showed biocompatibility and enhanced anti-TNBC activity.

Unfolding the Potency of Adenosine in Targeting Triple Negative Breast Cancer via Paclitaxel-Incorporated pH-Responsive Stealth Liposomes

Summary: In this study, a novel pH-sensitive liposome loaded with paclitaxel (PTX) and targeted with adenosine (ADN) was designed. The liposomes exhibited desirable particle size, drug encapsulation efficiency, and pH-responsive release. In cellular experiments, ADN-PEG-pH liposomes showed improved cellular uptake and apoptosis induction compared to free PTX. Moreover, ADN-PEG-pH liposomes demonstrated higher drug exposure and longer half-life than Intaxel. Overall, the development of this new drug delivery system is of great significance for TNBC treatment.

Lyophilization of Curcumin-Albumin Nanoplex with Sucrose as Cryoprotectant: Aqueous Reconstitution, Dissolution, Kinetic Solubility, and Physicochemical Stability

Summary: This study successfully developed a dry-powder form of the CUR-BSA nanoplex using sucrose as a cryoprotectant. Sucrose exhibited excellent cryoprotective activity, preserving the integrity of BSA and ensuring good aqueous reconstitution. While the aqueous reconstitution, CUR dissolution, and amorphous form of the nanoplex were minimally affected by elevated heat and humidity, the BSA payload decreased and protein aggregation increased after exposure. However, these adverse effects can be minimized by increasing the mass fraction of sucrose.

The nanoparticles can effectively accumulate in tumors, release drugs in response to intracellular pH and ROS levels, and amplify ROS generation with external laser irradiation to achieve extraordinary therapeutic efficacy. The treatment showed substantial tumor shrinkage and eradication with low toxicity in major organs, indicating great potential for tumor therapy.
