Some design considerations for cluster randomized trials with binary responses

Summary: This article discusses the optimal allocation of experimental units in a control versus intervention cluster randomized trial for binary responses. Optimal allocations are determined based on the well-known D-s- optimality criterion, and locally optimal designs, Bayesian optimal designs, and min-max optimal designs are proposed to address the issue of parameter dependency on design criterion. Efficiency analysis and numerical experiments, including a real data example, demonstrate a significant improvement in efficiency for the proposed designs compared to the commonly used balanced design.

Summary: In this article, the authors propose computationally efficient power and sample size procedures for stepped wedge cluster randomized trials (SW-CRTs) with multivariate outcomes. They derive the joint distribution of the intervention test statistics under a multivariate linear mixed model and provide an example using the intersection-union test for co-primary outcomes. The authors also prove that the multivariate linear mixed model leads to a more efficient treatment effect estimator compared to the univariate linear mixed model under certain assumptions.
