The role of VEGF in cancer-induced angiogenesis and research progress of drugs targeting VEGF

While it plays a crucial role in physiological homeostasis, it also supports tumor growth by providing the necessary oxygen and nutrients. Vascular endothelial growth factor (VEGF), a prominent pro-angiogenic factor, is a strategic target in therapeutic methods due to its involvement in tumor blood vessel formation. This article summarizes the molecular mechanisms of VEGF and its diverse role in cancer angiogenesis, as well as the current transformative strategies targeting VEGF to interfere with tumor growth.

In silico prediction of pharmacokinetic and physicochemical parameters using SwissADME suggested further optimization is required for these derivatives.

Doxorubicin-An Agent with Multiple Mechanisms of Anticancer Activity

Recent evidence suggests that DOX exhibits pleiotropic anticancer activity, including DNA damage, reactive oxygen species (ROS) production, apoptosis, senescence, autophagy, ferroptosis, and pyroptosis induction, as well as immunomodulation. This review aims to gather information on the anticancer mechanisms of DOX and its impact on the anti-tumor immune response, providing a rationale for the importance of DOX in modern cancer therapy.

Novel hydrazone-isatin derivatives as potential EGFR inhibitors: Synthesis and in vitro pharmacological profiling

Summary: Merging isatin and arylhydrazone moieties is an efficient strategy to obtain new potential anticancer derivatives. Fourteen hydrazone-isatin derivatives were synthesized and evaluated for their antiproliferative activity against the NCI-60 cancer cell line panel. Compound VIIIb was found to inhibit the epidermal growth factor receptor (EGFR) and possess drug-likeness properties, leading to decreased cell population in the G2/M phase and increased apoptosis, confirming its potential as a new proapoptotic compound.

Self-Adjuvanting Calcium-Phosphate-Coated Microcrystal-Based Vaccines Induce Pyroptosis in Human and Livestock Immune Cells

Summary: Successful vaccines require effective adjuvants to activate the innate immune system. Protein-coated microcrystals modified with calcium phosphate (CaP-PCMCs) represent a promising self-adjuvanting vaccine delivery system. In this study, we investigated the mechanism of action of CaP-PCMCs by studying their interaction with immune cells. We found that incubation of cells with CaP-PCMCs induced rapid cell death, accompanied by the activation of Caspase-1 and the release of interleukin-1β. These findings support CaP-PCMCs as a promising vaccine delivery system.

VEGF-B prevents excessive angiogenesis by inhibiting FGF2/FGFR1 pathway

Summary: Although the angiogenic effect of VEGF-B is not well understood, inhibitors of VEGF-B are being used in the treatment of neovascular diseases. In this study, we reveal a previously unrecognized function of VEGF-B as an endogenous inhibitor of angiogenesis by inhibiting the FGF2/FGFR1 pathway. We found that VEGF-B binds to FGFR1, induces FGFR1/VEGFR1 complex formation, suppresses FGF2-induced Erk activation, and inhibits FGF2-driven angiogenesis and tumor growth.

Abietic acid induced cell cycle arrest and subpopulation arrest in MCF-7, and also modulated the expression of apoptotic, proliferation, and oncogenic genes. Furthermore, abietic acid increased levels of antioxidants and showed potential as an anti-cancer agent in breast cancer therapy.
