The pathway of melatonin biosynthesis in common wheat (Triticum aestivum)

Natural variations of OsAUX5, a target gene of OsWRKY78, control the neutral essential amino acid content in rice grains

Summary: In this study, a previously unrecognized auxin influx carrier subfamily gene, OsAUX5, was identified to enhance the accumulation of essential amino acids (EAAs) in rice grains. Natural variations of OsAUX5 were found to contribute to the variation of grain EAAs among rice varieties. The introduction of OsAUX5 genotype from indica rice into japonica rice significantly increased EAA levels, suggesting a potential strategy for breeding EAA-rich rice.

Expanding our coverage: Strategies to detect a greater range of metabolites

Summary: The study of metabolites began several hundred years ago, but it gained momentum in the 20th century with a focus on single metabolites or simple metabolic pathways. The development of metabolomics and the use of high-resolution mass spectrometry now allows us to accurately annotate and quantify over 1000 plant metabolites. However, this falls short of the estimated number of metabolites in the plant kingdom. To improve metabolome coverage, three complementary strategies are reviewed, including the use of chemical libraries, molecular networking, and the integration of metabolomics and genetic variance. It is argued that using all three approaches in combination is the best way to address this challenge.

The combination of omics technologies allows for the identification of key genes, but the lack of efficient protocols for crop engineering remains a challenge. Hairy roots gene editing using CRISPR/Cas, coupled with omics analyses, provides a platform for rapid and precise functional analysis of genes. This article describes the requirements for efficient crop genome editing, focusing on recalcitrant legume transformation, and highlights the opportunities that gene editing in hairy roots offers for future crop improvement.

Rice metabolic regulatory network spanning the entire life cycle

Summary: This study identified 825 annotated metabolites using targeted and non-targeted metabolite profiling methods in rice samples from different tissues throughout the entire life cycle. A Rice Metabolic Regulation Network was constructed by combining these data with transcriptome data. The metabolites and co-expressed genes were divided into 12 clusters based on their accumulation patterns, providing a comprehensive metabolic profile of the rice life cycle. Novel transcription factors were identified using known regulatory genes and tissue specificity. The study demonstrates the importance of analyzing metabolome and transcriptome data in understanding agronomic traits.

Genome-wide association identifies a missing hydrolase for tocopherol synthesis in plants

Summary: The tocopherol biosynthetic pathway in plants is encoded by VTE genes, but many quantitative trait loci for seed total tocopherols do not have VTE genes, suggesting other activities are involved. A study on Arabidopsis seed tocopherols found a significant interval without VTE genes, which mapped to an uncharacterized, seed-specific, envelope-localized, alpha/beta hydrolase. Mutations in AtVTE7 and its maize ortholog ZmVTE7 significantly decreased seed and leaf total tocopherols, while overexpression of these genes increased leaf totaltocopherols. These findings suggest a model where VTE7 releases prenyl alcohols from chlorophyll biosynthetic intermediates for tocopherol biosynthesis.

Glycoside-specific metabolomics combined with precursor isotopic labeling for characterizing plant glycosyltransferases br

Integrative Analysis of Metabolome and Transcriptome Reveals the Role of Strigolactones in Wounding-Induced Rice Metabolic Re-Programming

Metabolomics-driven gene mining and genetic improvement of tolerance to salt-induced osmotic stress in maize

An LC-MS-based untargeted metabolomics approach identified 37 metabolite biomarkers related to SIOS tolerance, and 10 candidate genes significantly associated with SIOS tolerance and METO abundances. A citrate synthase, a glucosyltransferase, and a cytochrome P450 were validated to underlie genotype-METO-SIOS tolerance associations and improve the SIOS tolerance of elite maize inbred lines through additive effects.
