Recent advancements in single-cell multi-omics and genetic lineage tracing have revealed the transcriptional and phenotypical adaptation of endothelial cells to the injured microenvironment. These cells undergo transient changes in metabolism, mesenchymal properties, hematopoiesis, and pro-inflammatory responses. After the resolution of injury, endothelial cells revert back to their naïve state, preventing chronic dysfunction. Understanding how endothelial cells adapt to injury and contribute to repair and regeneration is important for maintaining a healthy microcirculation.
