Electrochemical Selenylation of Sulfoxonium Ylides for the Synthesis of gem-Diselenides as Antimicrobials against Fungi

Zhongnan Xu et al.

Summary: We report a metal-free, electrochemical method for the highly selective synthesis of valuable organoselenium compounds. By coupling alpha-keto sulfoxonium ylides with diselenides, the electrochemical selenylation reaction exhibits broad scope and functional group tolerance. Mechanistic studies reveal that the key C-Se bond is constructed using n-Bu4NI as an electrolyte and catalyst in the electrosynthetic protocol. The resulting alpha-keto gem-diselenides show excellent antimicrobial activity against Candida albicans, suggesting their potential as lead compounds for further exploration.

Electrochemical Organoselenium Catalysis for the Selective Activation of Alkynes: Easy Access to Carbonyl-pyrroles/oxazoles from N-Propargyl Enamines/Amides

Mrinmay Baidya et al.

Summary: Intramolecular electro-oxidative addition of enamines or amides to nonactivated alkynes was achieved to obtain carbonyl-pyrroles or-oxazoles from N-propargyl derivatives. Organoselenium was used as the electrocatalyst, functioning as a pi-Lewis acid to selectively activate the alkyne for successful nucleophilic addition. The synthetic strategy allows for a wide range of substrate scope with up to 93% yield. Several mechanistic experiments, including the isolation of a selenium-incorporated intermediate adduct, shed light on the electrocatalytic pathway.

Two-Component Redox Organocatalyst for Peptide Bond Formation

Handoko et al.

Summary: Peptides are important therapeutic drugs that can be synthesized automatically. However, the current peptide synthesis methods are not atom economical and require excessive coupling agents and protected amino acids. A recent study proposed a new organic catalyst that can be used in automated peptide synthesis, which reduces the amount of phosphine and eliminates the need for a dehydrating agent by utilizing a new catalytic cycle for amide bond formation.

Organoselenium Compounds as Acetylcholinesterase Inhibitors: Evidence and Mechanism of Mixed Inhibition

Amit Kumawat et al.

Summary: This study investigates the molecular mechanism of the mixed inhibition of AChE by ebselen and diphenyl diselenide (DPDSe) through systematic kinetic assays, intrinsic fluorescence tests, docking, and molecular dynamics (MD) simulations. The results show significant heterogeneity in the binding modes and allosteric hotspots for DPDSe on AChE, leading to non-competitive inhibition. Both ebselen and DPDSe bind strongly around the peripheral anionic site (PAS) and can enter the gorge through a side door, offering a promising novel entry point for AChE inhibitors.
