Novel CH25H+ and OASL+ microglia subclusters play distinct roles in cerebral ischemic stroke

Yueman Zhang et al.

Summary: Using single-cell RNA sequencing, researchers identified two microglia subclusters, CH25H(+) and OASL(+), that play important roles in ischemic stroke. CH25H(+) microglia exhibits increased phagocytosis and neuroprotective properties, while OASL(+) microglia is associated with neuroinflammation.

Microglia-mediated T cell infiltration drives neurodegeneration in tauopathy

Xiaoying Chen et al.

Summary: Extracellular deposition of amyloid-beta and intracellular accumulation of hyperphosphorylated, aggregated tau are characteristic hallmarks of Alzheimer's disease. The regional progression of brain atrophy highly correlates with tau accumulation, and the mechanisms of tau-mediated neurodegeneration remain elusive. This study reveals a tauopathy- and neurodegeneration-related immune hub involving activated microglia and T cell responses, which could serve as therapeutic targets for preventing neurodegeneration in Alzheimer's disease and primary tauopathies.

Microglia Exhibit Distinct Heterogeneity Rather than M1/M2 Polarization within the Early Stage of Acute Ischemic Stroke

Hongyu Ma et al.

Summary: This study used single-cell RNA sequencing to analyze the classification and phenotypic changes of microglia during the acute phase of ischemic stroke. The study found that microglia exhibited rich phenotypic diversity during the acute phase, and different subpopulations showed differences in inflammatory response and neurotrophic support. Furthermore, the study identified important cell-cell communication between microglia and other cell types. These findings contribute to identifying neuroprotective targets for early-stage ischemic stroke.

New Mechanisms and Targets of Subarachnoid Hemorrhage: A Focus on Mitochondria

Zeyu Zhang et al.

Summary: Spontaneous subarachnoid hemorrhage (SAH) is a subtype of hemorrhagic stroke that places a heavy burden on health care. Early brain injury (EBI) may lay the foundation for further delayed cerebral ischemia (DCI) development and poor outcomes. Mitochondrial dysfunction is considered a novel mechanism related to DCI and post-SAH outcomes.

The natural (poly)phenols as modulators of microglia polarization via TLR4/NF-κB pathway exert anti-inflammatory activity in ischemic stroke

Ruoqi Li et al.

Summary: This article discusses the role of inflammation in stroke and the therapeutic approach of modulating microglial phenotypes through the TLR4/NF-kappa B signaling pathway. It is found that dietary (poly)phenols have anti-inflammatory properties and provide protective effects on ischemic stroke.

Inhibition of caspase-1-mediated inflammasome activation reduced blood coagulation in cerebrospinal fluid after subarachnoid haemorrhage

Yuanjian Fang et al.

Summary: This study investigates the role of caspase-1-mediated inflammasome activation on extrinsic blood coagulation in cerebrospinal fluid (CSF) after subarachnoid hemorrhage (SAH). The results demonstrate that inhibiting caspase-1 with VX-765 can attenuate neuroinflammation and blood coagulation, leading to improved neurological outcomes and memory capacity in SAH rats. These findings suggest that VX-765 may be a potential treatment for SAH-related neuroinflammatory and hemostatic disorders.

Osteopontin in post-subarachnoid hemorrhage pathologies

Reona Asada et al.

Summary: The rupture of intracranial aneurysms leading to subarachnoid hemorrhage is a difficult condition to treat. After surgical removal of the ruptured aneurysms, other complications such as delayed cerebral ischemia and chronic hydrocephalus may occur. Osteopontin (OPN) plays a crucial role in regulating these complications.

MFG-E8 attenuates inflammation in subarachnoid hemorrhage by driving microglial M2 polarization

Yong-Yue Gao et al.

Summary: The study found that rhMFG-E8 can inhibit brain edema, improve neurological function, reduce inflammatory factor levels, and promote microglial transformation to the M2 phenotype. However, inhibition of MFG-E8 and integrin beta 3 via specific siRNA, as well as the STAT3 inhibitor Stattic, can abolish the effects of MFG-E8 on anti-inflammation and M2 phenotype polarization.
