Specialized Pro-Resolving Mediators as Resolution Pharmacology for the Control of Pain and Itch

Ru-Rong Ji

Summary: Specialized pro-resolving mediators (SPMs) are endogenous lipid mediators synthesized from omega-3 polyunsaturated fatty acids during inflammation. Synthetic SPMs have shown potent inflammation-resolving effects and analgesia in preclinical models, including rodent models of different types of pain. SPMs act through multiple G protein-coupled receptors expressed by immune cells, glial cells, and neurons, and their loss can impair pain resolution. SPMs also have an emerging role in itch control, and targeting SPMs or SPM receptors pharmacologically could lead to novel therapeutics for pain and itch.

Serum lipid mediator profiles in COVID-19 patients and lung disease severity: a pilot study

Pilar Irun et al.

Summary: COVID-19 patients show increased levels of pro-inflammatory lipid mediators, while the role of anti-inflammatory mediators is less explored. This study analyzed the lipid mediator metabolome in COVID-19 patients and found significant increases in several inflammation-related substances in severe cases. These findings highlight the complexity of the inflammatory response in COVID-19 and suggest the need for further research to identify new therapeutic targets.

Pro-inflammatory and pro-resolving lipid mediators of inflammation in HIV: effect of aspirin intervention

Jesmond Dalli et al.

Summary: People with HIV have an increased risk of cardiovascular disease compared to HIV-negative individuals. In this study, we found that HIV-positive individuals have lower levels of pro-inflammatory lipid mediators and higher levels of pro-resolving lipid mediators compared to HIV-negative individuals. We also observed that aspirin treatment in HIV-positive individuals reduced the levels of pro-inflammatory lipid mediators and upregulated certain pro-resolving lipid mediators.

Blueberry intake elevates post-exercise anti-inflammatory oxylipins: a randomized trial

David C. Nieman et al.

Summary: This study aimed to investigate the effects of blueberry supplementation on muscle soreness, damage, and inflammation resolution in untrained adults after intense exercise. The results showed that daily supplementation with freeze-dried blueberries significantly increased urinary phenolic levels and reduced inflammation. However, there were no significant differences between the blueberry and placebo groups in exercise performance and muscle soreness.

The effect of low-dose aspirin on aspirin triggered lipoxin, interleukin 1 beta, and prostaglandin E2 levels in periapical fluid: a double-blind randomized clinical trial

Elham Khoshbin et al.

Summary: This randomized clinical trial evaluated the effect of low-dose aspirin on the levels of pro-inflammatory mediators and pro-resolving mediators in periapical fluid of necrotic teeth with large lesions. The results showed that low-dose aspirin can reduce the production of pro-inflammatory mediators such as PGE2 and IL-1β, while increasing the production of pro-resolving mediators such as ATL.

Enriched Marine Oil Supplement Increases Specific Plasma Specialized Pro-Resolving Mediators in Adults with Obesity

Abrar E. Al-Shaer et al.

Summary: A marine oil supplement can increase the concentrations of specific specialized pro-resolving mediators (SPMs) and their metabolic intermediates in adults with obesity, which may be beneficial for controlling inflammation in obesity. However, further studies are needed to determine if increased concentrations of specific SPMs can control the resolution of inflammation in humans with obesity.

Topical Administration of a Marine Oil Rich in Pro-Resolving Lipid Mediators Accelerates Wound Healing in Diabetic db/db Mice through Angiogenesis and Macrophage Polarization

Imelda Ontoria-Oviedo et al.

Summary: Impaired wound healing in patients with type 2 diabetes is characterized by chronic inflammation. Specialized pro-resolving lipid mediators (SPMs) are bioactive molecules that regulate inflammation. This study found that LIPINOVA(R), a nutritional supplement rich in SPMs and EPA, promotes wound healing by reducing inflammation and inducing angiogenesis and macrophage polarization.

PUFA Supplementation and Heart Failure: Effects on Fibrosis and Cardiac Remodeling

Francesca Oppedisano et al.

Summary: Dietary supplementation with n-3 polyunsaturated fatty acids (PUFAs) has shown positive effects on heart failure by reducing inflammation, protecting the heart, and lowering the risk of hospitalization and death. Studies suggest that n-3 PUFAs play a role in inhibiting cardiac fibrosis and improving cardiac function in HF patients.
