A Hairpin Motif in the Amyloid-& beta; Peptide Is Important for Formation of Disease-Related Oligomers

Mohammed Khaled et al.

Summary: The formation of Aβ oligomers is dependent on the presence of a specific β-hairpin motif in the peptide sequence. Oligomers initially grow spherically but start to form extended linear aggregates at larger oligomeric states. The population of extended oligomers can be increased by introducing an intramolecular disulfide bond, while truncating a β-strand-forming segment decreases the oligomer population and removes the formation of extended oligomers.

β-Turn exchanges in the α-synuclein segment 44-TKEG-47 reveal high sequence fidelity requirements of amyloid fibril elongation

Emil Dandanell Agerschou et al.

Summary: The study shows that the formation of turns in the αS protein significantly affects its fibril formation kinetics. Variants with faster beta 1'- or beta 2'-turn formation tend to aggregate more rapidly, while variants with beta 1-turn formation show slower aggregation compared to wild-type. Furthermore, turn exchanges alter fibril morphology and affect the efficiency of fibril elongation.

Structural insights into α-synuclein monomer-fibril interactions

Pratibha Kumari et al.

Summary: This study investigated the interaction between monomeric alpha-Syn and its fibrillar form using NMR and electron paramagnetic resonance spectroscopy, revealing that intermolecular interactions reduce intramolecular contacts in monomeric alpha-Syn, leading to further unfolding of its intrinsically disordered states and critically contributing to the aggregation kinetics of alpha-Syn during secondary nucleation.
