Preliminary Studies of Antimicrobial Activity of New Synthesized Hybrids of 2-Thiohydantoin and 2-Quinolone Derivatives Activated with Blue Light

Summary: Thiohydantoin and quinolone derivatives with broad-spectrum medical applications have been synthesized and analyzed for their antimicrobial properties. The effect of blue light irradiation on these compounds was also investigated. Preliminary results showed that some of the hybrid compounds exhibited bacteriostatic activity towards Gram-positive and Gram-negative bacteria, and this effect was enhanced by blue light activation.

Synthesis, Crystal Structures, Lipophilic Properties and Antimicrobial Activity of 5-Pyridylmethylidene-3-rhodanine-carboxyalkyl Acids Derivatives

Summary: The constant increase in resistance of pathogenic bacteria to commonly used drugs necessitates the search for new antibacterial substances. In this study, a series of rhodanine-3-carboxyalkyl acid derivatives containing pyridinyl moiety were obtained and tested for their antibacterial and antifungal activities. The results showed activity against Gram-positive bacteria while they were inactive against Gram-negative bacteria and yeast. Crystal structures and lipophilicity analysis revealed that derivatives with 1.5 NS electrostatic interactions exhibited the highest biological activity.

Molecular Insights into an Antibiotic Enhancer Action of New Morpholine-Containing 5-Arylideneimidazolones in the Fight against MDR Bacteria

Summary: New morpholine-containing compounds were explored as potential antibiotic adjuvants against Gram-positive and Gram-negative bacteria, with some compounds showing promising results in overcoming antibiotic resistance. Molecular modelling and experimental testing confirmed their mechanisms of action, while also demonstrating significant MDR-reversal dual action and inhibition of efflux pumps in resistant bacteria.

An insight into the structure of 5-spiro aromatic derivatives of imidazolidine-2,4-dione, a new group of very potent inhibitors of tumor multidrug resistance in T-lymphoma cells

Summary: A series of newly synthesized compounds were explored for their potential as potent inhibitors of cancer multidrug resistance efflux pumps. Particularly, the spirofluorene derivatives with amine alkyl substituents at position 1 showed the strongest P-glycoprotein inhibitory activity in MDR T-lymphoma cells, with compounds 7 and 12 being 100-fold more potent than verapamil. Crystallography-supported pharmacophore-based SAR analysis suggested specific structural properties contributing to this excellent cancer MDR-inhibitory action.

Efflux Pump Mediated Antimicrobial Resistance by Staphylococci in Health-Related Environments: Challenges and the Quest for Inhibition

Summary: The increasing antimicrobial resistance in staphylococcal bacteria poses a major health threat globally, with decreased discovery of new antibiotics and increased use of sanitizers exacerbating the issue. Combining efflux pump inhibitors with current antibiotics to combat resistant staphylococci is a promising strategy. Future research on high-resolution structures of staphylococcal efflux pumps could lead to the development of more effective inhibitors for clinical use.
