Challenges in Expression and Purification of Functional Fab Fragments in E. coli: Current Strategies and Perspectives

Rucha S. Patil et al.

Summary: Escherichia coli is the preferred host for biotherapeutics production due to its ease of cloning, scale-up, high yields, and cost-effective cultivation. However, it faces difficulties in producing biologically active therapeutics such as Fab fragments. This paper outlines recent improvements in upstream and downstream unit operations for producing Fab fragments in E. coli, including the coexpression of molecular chaperones and strain engineering for post-translational modifications.
