SARS-CoV-2 Detection and Culture in Different Biological Specimens from Immunocompetent and Immunosuppressed COVID-19 Patients Infected with Two Different Viral Strains

Maria Cassia Mendes-Correa et al.

Summary: By analyzing samples from 98 immunocompetent and 25 immunosuppressed individuals over five weeks, it was found that SARS-CoV-2 detection was highest in nasopharyngeal swabs and the duration of viral shedding varied among different clinical specimens and individuals. Prolonged shedding of potentially infectious virus primarily occurred in immunosuppressed individuals and virus replication was observed in a minority of subjects in vitro.

Gustavo Roncoli Reigado et al.

Summary: The study focused on fusing Trichoderma reesei superoxide dismutase with transactivator of transcription (TAT) of HIV-1 and abalone (Ab) peptides to enhance antioxidant protection on skin cells. The results revealed that the recombinant Ab-TrSOD was more effective in reducing oxidative stress compared to TrSOD without cell-penetrating peptide, and showed no cytotoxic effects on cells at the concentrations tested.

Summary: This study investigated the effectiveness of using a combination of specific antigens and adjuvants through intranasal administration in inducing immune responses. The results showed that this strategy was able to induce both mucosal and systemic immune responses. Additionally, lipid adjuvants were found to enhance the vaccine effect.

From a recombinant key antigen to an accurate, affordable serological test: Lessons learnt from COVID-19 for future pandemics

Summary: This study discusses the development of an accurate and affordable COVID-19 antibody test, including the production of a recombinant protein antigen and the validation of an enzyme-linked immunosorbent assay (ELISA). The test showed high accuracy with 98.6% specificity and 95% sensitivity, and dried blood spots collected on test strips could replace conventional plasma/serum samples. The estimated cost was low, and the entire process could be completed in four months. This research is important for reliable and affordable seroepidemiological surveillance during pandemics.

Neutralizing antibodies to SARS-CoV-2 variants of concern including Delta and Omicron in subjects receiving mRNA-1273, BNT162b2, and Ad26.COV2.S vaccines

George Fei Zhang et al.

Summary: SARS-CoV-2 vaccines show varying levels of effectiveness against different variants, with Omicron being the most challenging due to its high number of mutations. Two-dose mRNA-1273 and BNT162b2 vaccines provide robust neutralizing antibodies against multiple variants, but their effectiveness wanes after 6 months. One dose of Ad26.COV2.S vaccine induces weaker antibodies, particularly against Omicron. Booster shots with mRNA-based vaccines are recommended 5 months after full immunization, and mRNA-based vaccines are suggested 2 months after Ad26.COV2.S vaccination.

Promotion of neutralizing antibody-independent immunity to wild-type and SARS-CoV-2 variants of concern using an RBD-Nucleocapsid fusion protein

Julia T. Castro et al.

Summary: This study generated a chimeric protein (SpiN) that can elicit specific T cell responses and provide strong protection against SARS-CoV-2 infection. The results suggest that vaccine formulations that induce effector T cells specific for the nucleocapsid antigen and receptor binding domain of SARS-CoV-2 can improve COVID-19 vaccines and potentially overcome immune escape by variants of concern.

Dynamics of SARS-CoV-2 Variants of Concern in Vaccination Model City in the State of Sao Paulo, Brazil

Svetoslav Nanev Slavov et al.

Summary: Brazil, with one of the highest rates of COVID-19 infections and deaths, has implemented a successful vaccination program that reduced mortality and morbidity. The study conducted in the town of Serrana revealed that the vaccine campaign significantly reduced deaths and morbidity during the Gamma and Delta variants of concern (VOC). However, the Omicron VOC showed rapid substitution and morbidity rates despite vaccination.

IgA dominates the early neutralizing antibody response to SARS-CoV-2

Delphine Sterlin et al.

Summary: The study found that humoral immune responses to SARS-CoV-2 infection are dominated by IgA antibodies, with specific IgA plasmablasts rapidly expanding shortly after symptom onset and peaking in the third week of the disease. Virus-specific antibody responses include IgG, IgM, and IgA, with IgA playing a greater role in virus neutralization compared to IgG.

Summary: Lipid nanoparticle (LNP)-encapsulated nucleoside-modified mRNA vaccines have intrinsic adjuvant activity that enhances durable and protective antibody responses. Compared to the widely used MF59-like adjuvant AddaVax, this LNP formulation outperforms and relies on specific components and cytokine induction for its adjuvant activity.

Summary: IgA is unique among antibodies as it can survive on mucosal surfaces and play a critical role in preventing enteric infections, although its ability to activate effector functions is limited. Its distinct antigen-binding repertoire is shaped by local cues and the microbiota, crucial for regulating intestinal bacteria.

Summary: Research demonstrates that both humoral and cellular immunity play a role in clearing SARS-CoV-2, and convalescent mice or mice vaccinated with mRNA are protected from infection with both the wild type virus and the B.1.351 variant. This protection is mainly mediated by antibody response rather than cellular immunity.
