Pharmaceutical pollution of the world's rivers

John L. Wilkinson et al.

Summary: Environmental exposure to active pharmaceutical ingredients (APIs) can have negative effects on ecosystems and human health. A global-scale study of API pollution in rivers highlights the highest concentrations in sub-Saharan Africa, south Asia, and South America, primarily in low- to middle-income countries with poor wastewater and waste management infrastructure. The most frequently detected APIs include carbamazepine, metformin, and caffeine, with some concentrations exceeding safe levels for aquatic organisms. Pharmaceutical pollution poses a global threat to environmental and human health and has implications for achieving the United Nations Sustainable Development Goals.

Towards the design of active pharmaceutical ingredients mineralizing readily in the environment

Neele Puhlmann et al.

Summary: Active pharmaceutical ingredients (APIs), their metabolites, and transformation products (TPs) are globally present in the environment, posing risks to both environmental and human health. The concept of Benign by Design (BbD) in designing APIs for mineralization in the environment is a promising approach to address this challenge, but its implementation in the industrial API discovery process has not been discussed. The optimization phase within drug discovery appears to be the most suitable stage to integrate environmental considerations, with the feasibility of BbD depending on the availability of suitable tools and methods as well as incentives for research and development.
