Senescence mechanisms and targets in the heart

Maggie S. Chen et al.

Summary: Cellular senescence is an irreversible cell cycle arrest associated with ageing. It plays a critical role in the pathophysiology of cardiovascular diseases, such as atherosclerosis, myocardial infarction, and cardiac fibrosis. Understanding the molecular mechanisms of cellular senescence and its consequences is important for developing new therapeutic approaches to treat CVD.

Inflammation, Aging, and Cardiovascular Disease JACC Review Topic of the Week

Luca Liberale et al.

Summary: Aging and inflammation both play key roles in cardiovascular and cerebrovascular diseases, recognizing the concept of inflamm-aging can lead to new therapeutic targets and options for the growing aging population. This review discusses recent scientific advances in understanding the pathways of inflamm-aging and its potential implications in clinical practice.

Cardiovascular Inflammaging: Mechanisms and Translational Aspects

Maria Luisa Barcena et al.

Summary: Aging is a major risk factor for chronic diseases including cancer, type 2 diabetes, dementia, and cardiovascular diseases. The underlying cellular mechanisms involve complex processes such as telomere shortening and chronic low-grade inflammation. This review focuses on the molecular mechanisms and translational aspects of cardiovascular aging-related inflammation.

Effect of Hypoxia on Pulmonary Endothelial Cells from Bleomycin-Induced Pulmonary Fibrosis Model Mice

Daisuke Akahori et al.

Summary: The study showed that under hypoxic conditions, specific protein expression in pulmonary endothelial cells was affected, leading to enhanced fibrogenesis-related damage, and this phenomenon was more pronounced in patients with pulmonary fibrosis.

Inflammation and cardiovascular diseases: lessons from seminal clinical trials

Luca Liberale et al.

Summary: Canakinumab Anti-inflammatory Thrombosis Outcome Study (CANTOS) and Colchicine Cardiovascular Outcomes Trial (COLCOT) were the first two randomized clinical trials to demonstrate the effectiveness of specific anti-inflammatory treatments in cardiovascular prevention, confirming the involvement of specific inflammatory pathways in atherosclerotic cardiovascular disease. Other trials targeting multiple inflammasome-independent pathways failed to show benefit, highlighting the significance of balancing the risk of impaired host defense and infections with the prevention of major adverse cardiovascular events in patients with residual inflammatory risk.

Lacidipine Ameliorates the Endothelial Senescence and Inflammatory Injury Through CXCR7/P38/C/EBP-β Signaling Pathway

Xing Liu et al.

Summary: Lacidipine treatment lowers blood pressure in SHR by increasing CXCR7 expression and suppressing P38 and C/EBP-beta. It enhances EC cell viability and function under oxidative stress, cell senescence, and inflammatory activation via the CXCR7/P38 signaling pathway.

SIRT6 Protects Smooth Muscle Cells From Senescence and Reduces Atherosclerosis

Mandy O. J. Grootaert et al.

Summary: The study reveals that SIRT6 protein expression in VSMCs is reduced in human and mouse plaque VSMCs and is positively regulated by CHIP. SIRT6 plays a role in regulating telomere maintenance and VSMC lifespan, inhibiting atherosclerosis through its deacetylase activity.

Inflammation in Atherosclerosis-No Longer a Theory

Peter Libby

Summary: Inflammation is closely linked to atherosclerosis, and recent clinical trials have shown the effectiveness of anti-inflammatory interventions in improving outcomes for patients with the disease. Further research is needed to optimize these interventions and improve patient selection.

Roxithromycin attenuates bleomycin-induced pulmonary fibrosis by targeting senescent cells

Xuan Zhang et al.

Summary: This study investigated the potential of roxithromycin in treating pulmonary fibrosis by targeting senescent cells. Results showed that roxithromycin could selectively kill senescent cells, inhibit senescent cell-induced lung fibroblast activation, attenuate lung injury, inflammation, and fibrosis, and achieve these effects by inhibiting the NOX4 pathway.

CD38 deficiency alleviates Ang II-induced vascular remodeling by inhibiting small extracellular vesicle-mediated vascular smooth muscle cell senescence in mice

Lu Gan et al.

Summary: CD38 deficiency alleviates vascular remodeling induced by Ang II, primarily affecting VSMCs rather than lymphocytes. CD38 deficiency or NAD supplementation mitigates vascular senescence induced by Ang II. The protective effects of CD38 deficiency on VSMC senescence are related to restoration of lysosome dysfunction.
