Frequency of food allergy in Europe: An updated systematic review and meta-analysis

Giulia C. Spolidoro et al.

Summary: The latest estimates of food allergy prevalence and trends in Europe show an increase in self-reported food allergy and sensitization rates. This may be due to a real increase in the prevalence, increased awareness, increased evaluation of food types, or increased number of studies. Future studies need to use standardized methods, including double-blinded placebo-controlled food challenge, to minimize potential biases.

Rewiring the altered tryptophan metabolism as a novel therapeutic strategy in inflammatory bowel diseases

Chloe Michaudel et al.

Summary: This study identified a new mechanism linking tryptophan metabolism to intestinal inflammation and inflammatory bowel diseases (IBDs). Bringing back xanthurenic acid (XANA) and kynurenic acid (KYNA) has protective effects involving Aryl hydrocarbon Receptor (AhR) activation and the rewiring of cellular energy metabolism in intestinal epithelial cells and CD4(+) T cells. Manipulating the endogenous metabolic pathway with aminoadipate aminotransferase (AADAT) may provide new therapeutic strategies for correcting tryptophan metabolism alterations in IBD.

The food additive titanium dioxide hinders intestinal production of TGF-& beta; and IL-10 in mice, and long-term exposure in adults or from perinatal life blocks oral tolerance to ovalbumin

Bruno Lamas et al.

Summary: Food hypersensitivities are on the rise in industrialized countries, and foodborne nanoparticles (NPs) are suspected to play a role. This study investigated the effects of food-grade titanium dioxide (fg-TiO2) on oral tolerance (OT) to food proteins in mice. The results showed that long-term exposure to fg-TiO2 altered the anti-inflammatory cytokine profile and led to OT failure, regardless of the timing of exposure.

Epithelial barrier hypothesis: Effect of external exposome on microbiome and epithelial barriers in allergic disease

Zeynep Celebi Sozener et al.

Summary: Environmental exposure plays a major role in the development of allergic diseases, including climate change, pollution, changes and loss of biodiversity, etc. The epithelial barrier hypothesis provides a mechanistic explanation of how these factors contribute to the increase in allergic and autoimmune diseases. Measures should be taken to mitigate environmental threats and decrease allergic diseases.

Impact of intracellular innate immune receptors on immunometabolism

Wei-Chun Chou et al.

Summary: Immunometabolism plays a crucial role in regulating immune cell functions, with the interaction between PI3K-AKT-mTOR and LKB1-AMPK signaling pathways being essential for modulating cell metabolism. Intracellular innate immune sensors/receptors have a significant impact on metabolic pathways, highlighting the need for further investigation into their mechanisms of interaction.

The Development of Early Life Microbiota in Human Health and Disease

Hanying Lv et al.

Summary: The colonization of the human microbiota in early life has significant implications for long-term health, particularly in relation to metabolic, allergic, and autoimmune diseases. Understanding the development process and factors influencing the gut microbiota, as well as its interaction with the host's physiological systems, is crucial for deciphering the mechanisms underlying diseases associated with the early life gut microbiome. Microbiota-targeted therapeutic methods show promising effects in treating these diseases.

Intestinal Barrier Permeability in Allergic Diseases

Monika Niewiem et al.

Summary: The role of intestinal permeability in food allergies is crucial, as it affects the immune system and the integrity of the epithelial barrier. However, the understanding of this process is still limited, and further research is needed to explore relevant indicators.

Intestinal epithelium in early life

Lauren C. Frazer et al.

Summary: Rapid development of the fetal and neonatal intestine is necessary for growth and protection. In utero, the fetus receives nutrition and is protected, allowing the intestine to develop in a quiescent environment. After birth, the intestine needs to provide defense and nutrition, which is achieved through the development of specialized epithelial cells from intestinal stem cells. Interrupting these homeostatic processes can lead to intestinal diseases, such as necrotizing enterocolitis.

Toxic effects of TiO2 NPs in the blood-milk barrier of the maternal dams and growth of offspring

Liyang Yao et al.

Summary: This study revealed that TiO2 NPs can induce toxicity in maternal dams, such as pathological damages to mammary gland tissues, and transfer to offspring via breastfeeding, causing abnormal development in infants. Administration of vitamin E could rescue the toxic effects induced by TiO2 NPs.

Safety assessment of titanium dioxide (E171) as a food additive

Maged Younes et al.

Summary: This opinion presents an updated safety assessment of the food additive titanium dioxide (E 171) based on new scientific evidence. Studies indicated that while particles smaller than 30 nm may have limited relevance to E 171 safety assessment, general and organ toxicity were not observed at the tested doses. Adverse effects were indicated in terms of potential immunotoxicity and inflammation with E 171 and potential neurotoxicity with TiO2 NPs.

The Importance of Metabolism for Immune Homeostasis in Allergic Diseases

Juan Rodriguez-Coira et al.

Summary: The metabolic status of T cells and macrophages is closely related to severe phenotypes of chronic inflammation, including allergic inflammation. Metabolic changes in immune cells play a crucial role in their inflammatory or regulatory responses, and understanding these alterations could help restore and modulate immune cell functions. This link between energy metabolism and inflammation can be studied using various models and analytical approaches, from classic immunological studies to integrated analysis of metabolomics, transcriptomics, and proteomics.

Cezmi A. Akdis

Summary: The increase in allergic, autoimmune, and chronic diseases may be related to epithelial barrier-damaging agents associated with industrialization, urbanization, and modern life. These diseases are more prevalent in industrialized countries and are on the rise in developing countries, affecting over one billion people globally.
