Magnetic self-assembly of 3D multicellular microscaffolds: A biomimetic brain tumor-on-a-chip for drug delivery and selectivity testing

Attilio Marino et al.

Summary: In recent years, there has been an increasing demand for highly predictive brain cancer models to test new anticancer compounds and experimental therapeutic approaches. Realistic in vitro brain tumor-on-a-chip platforms can lead to more accurate selection of potential drugs, reducing the economic and ethical issues associated with unsuccessful in vivo studies.

Evaluation of Proton-Induced DNA Damage in 3D-Engineered Glioblastoma Microenvironments

Qais Akolawala et al.

Summary: A novel 3D-engineered scaffold inspired by the geometry of brain blood vessels was designed to assess the effects of proton therapy on glioblastoma (GBM) cells. The results showed a higher DNA double-strand breakage in 3D-cultured cells compared to 2D cell monolayers, indicating potential differences in radioresistance of GBM cells between different culture configurations. These biomimetic-engineered 3D scaffolds provide a benchmark tool for evaluating the effects of proton therapy on 3D GBM cell networks and other types of cancer cells.

Radioresistance and Transcriptional Reprograming of Invasive Glioblastoma Cells

Zili Tang et al.

Summary: This study reveals that invasive tumor cells in glioblastoma exhibit high radioresistance and are closely associated with patient prognosis. Transcriptome analysis identifies the gene expression profile of invasive cells and validates the involvement of key pathways. Inhibition of specific signaling pathways and the use of carbon ion therapy can enhance the sensitivity of invasive cells to radiation treatment.

Association between vascular endothelial growth factor-mediated blood-brain barrier dysfunction and stress-induced depression

Hitomi Matsuno et al.

Summary: Several lines of evidence suggest that stress-induced neurovascular dysfunction and increased blood-brain barrier permeability are important pathological links between stress and psychiatric disorders. This study demonstrates the crucial role of vascular endothelial growth factor in stress-induced blood-brain barrier dysfunction and depressive-like behavior development.

Engineered cell culture microenvironments for mechanobiology studies of brain neural cells

Lucia Castillo Ransanz et al.

Summary: The biomechanical properties of the brain microenvironment play a critical role in brain development and neural functioning. However, these properties are often overlooked in current cell culture systems. Thus, there is a need to develop more representative in vitro brain models to study the mechanical aspects of the neural microenvironment.

3D-Engineered Scaffolds to Study Microtubes and Localization of Epidermal Growth Factor Receptor in Patient-Derived Glioma Cells

Nastaran Barin et al.

Summary: A major obstacle in glioma research is the lack of in vitro models that can retain cellular features of glioma cells in vivo. This study developed a 3D-engineered scaffold as a cell culture model system to study patient-derived glioma cells. Compared to conventional 2D cell cultures, the 3D-engineered scaffolds more closely resemble in vivo glioma cellular features and allow better monitoring of individual cells and intracellular trafficking.

Summary: This study provides an in-depth analysis of the cellular composition and microenvironment of human glioblastoma, revealing its structural and biological characteristics. The critical association between tumor cells and neo-vascular niche, as well as the relationship between T cell infiltration and tumor proliferation, were highlighted. The research also discovered distinct cell phenotypes in tumor nests and surrounding tissues.

Summary: The study introduced a novel method to produce mesenchymal cell-derived matrices (CDMs) aimed at mimicking the fibrotic tumor microenvironment, showing that they closely mimic the composition, structure, and mechanical properties of human fibrotic ECM. CDMs have highly reproducible composition and tunable mechanical properties, and exhibit increased doxorubicin resistance in cancer cells following chemotherapeutic treatment in comparison to 3D culture in collagen hydrogels.

3D In Vitro Models for Investigating the Role of Stiffness in Cancer Invasion

Auxtine Micalet et al.

Summary: This review discusses the role of the physical microenvironment in cancer invasion, finding that increased matrix stiffness may promote cancer invasion, but there are also cases where the opposite occurs. Experimental approaches and interpretations vary, affecting the invasion of cancer differently. The findings can guide future research methods and provide a standardized experimental protocol.
