Integrated 5-hydroxymethylcytosine and fragmentation signatures as enhanced biomarkers in lung cancer

Xinlei Hu et al.

Summary: This study successfully constructed a diagnostic model for lung cancer using the 5hmC signal and fragmentation profiles in 5hmC cfDNA data, which can effectively distinguish lung cancer patients from healthy individuals. The models showed good performance in different lung cancer subtypes and stages, and were associated with patient survival rates. The results demonstrate the significant application value of clinical diagnostic models based on cfDNA 5hmC data.

LRP1B: A Giant Lost in Cancer Translation

Catarina Principe et al.

Summary: LRP1B is a member of the LDLR protein family and is considered a potential tumor suppressor in human cancer. However, limitations exist in studying LRP1B due to its huge size. New evidence suggests an increasing relevance of LRP1B in cancer.

Global cancer statistics 2020: GLOBOCAN estimates of incidence and mortality worldwide for 36 cancers in 185 countries

Hyuna Sung et al.

Summary: The global cancer burden in 2020 saw an estimated 19.3 million new cancer cases and almost 10.0 million cancer deaths. Female breast cancer surpassed lung cancer as the most commonly diagnosed cancer, while lung cancer remained the leading cause of cancer death. These trends are expected to rise in 2040, with transitioning countries experiencing a larger increase compared to transitioned countries due to demographic changes and risk factors associated with globalization and a growing economy. Efforts to improve cancer prevention measures and provision of cancer care in transitioning countries will be crucial for global cancer control.

Targeted therapy for LIMD1-deficient non-small cell lung cancer subtypes

Kathryn Davidson et al.

Summary: The loss of the tumour suppressor gene LIMD1 is an early event in lung oncogenesis, affecting approximately 45% of non-small cell lung cancers. A drug repurposing screen identified PF-477736 as selectively targeting LIMD1-deficient cells, inducing cell death through apoptosis. PF-477736 showed effectiveness in treating LIMD1(-/-) tumors in xenograft models, presenting a potential new therapeutic subgroup for critical unmet needs.

Alterations of 5-hydroxymethylation in circulating cell-free DNA reflect molecular distinctions of subtypes of non-Hodgkin lymphoma

Brian C. -H. Chiu et al.

Summary: This study identified differential genome-wide 5hmC profiles in circulating cell-free DNA between DLBCL and FL, suggesting a potential non-invasive approach for distinguishing the two lymphomas. Moreover, the 5hmC modification levels in signature genes may provide insights into predicting risk of all-cause mortality in patients.

Screening for Lung Cancer CHEST Guideline and Expert Panel Report

Peter J. Mazzone et al.

Summary: Evidence suggests that low-dose CT screening for lung cancer can result in a favorable balance of benefit and harms. Factors such as the selection of screen-eligible individuals, the quality of imaging and image interpretation, the management of screen-detected findings, and the effectiveness of smoking cessation interventions can impact this balance.
