Translatome analysis reveals microglia and astrocytes to be distinct regulators of inflammation in the hyperacute and acute phases after stroke

Victoria G. Hernandez et al.

Summary: Neuroinflammation, as a significant feature of ischemic stroke, plays a critical role in its progression. Understanding the cellular signaling pathways that initiate and propagate neuroinflammation is important for developing immunomodulatory stroke therapies. This study reveals that microglia and astrocytes respond differently to stroke, with microglia adopting an inflammatory profile and astrocytes showing stress response genes and pro-inflammatory pathways. The findings provide valuable insights into the mechanisms of neuroinflammation after stroke.

Biological Mechanism of Sex Difference in Stroke Manifestation and Outcomes

Wi-Sun Ryu et al.

Summary: Female patients with acute ischemic stroke have greater stroke severity and higher risk of early neurologic deterioration, leading to lower functional independence at 3 months compared to male patients. These differences may be attributed to variations in infarct locations and their impact on neurological outcomes.

Reactive astrocytes acquire neuroprotective as well as deleterious signatures in response to Tau and Aβ pathology

Zoeb Jiwaji et al.

Summary: Alzheimer's disease affects astrocytes, and both A beta and Tau pathology induce changes in astrocytes. A beta mainly influences the expression of AD risk genes, while both pathologies induce age-dependent changes and have overlapping features with astrocytes found in post-mortem AD patients. Both A beta and Tau pathology alter the characteristics of astrocytes, including repression of bioenergetic and translation machinery, induction of inflammation pathways, and expression of protein degradation/proteostasis genes. Astrocyte-specific expression of Nrf2, a protective factor, can recapitulate elements of these astrocyte changes and reduce A beta deposition and phospho-tau accumulation. This Nrf2 expression also improves brain-wide transcriptional deregulation, cellular pathology, neurodegeneration, and behavioral/cognitive deficits.

Female-specific neuroprotection after ischemic stroke by vitronectin-focal adhesion kinase inhibition

Cuihong Jia et al.

Summary: Blood vitronectin (VTN) leaking into the brain after stroke exacerbates tissue loss by increasing IL-6 expression in female mice. Inhibiting focal adhesion kinase (FAK) reduces brain injury size, inflammation, and motor dysfunction in female, but not male, mice. Selective gene deletion of FAK in astrocytes reduces IL-6 expression and mitigates infarct size and inflammation. VTN signaling can be targeted to mitigate harmful inflammation in female stroke patients.

Insulin regulates neurovascular coupling through astrocytes

Ana M. Fernandez et al.

Summary: Insulin receptors in astrocytes play a role in neurovascular coupling by modulating glucose uptake and angiogenesis. Lack of insulin receptors in astrocytes leads to uncoupling of brain blood flow with glucose uptake, disturbed mitochondrial function, and abnormal angiogenesis. Treatment with the antioxidant N-acetylcysteine can normalize these effects.

Microglial metabolic disturbances and neuroinflammation in cerebral infarction

Haruna Takeda et al.

Summary: Cerebral ischemia/reperfusion injury triggers the activation of microglia and infiltration of circulating immune cells, which contribute to the pathogenesis of cerebral infarction. Metabolic alterations in immune cells, particularly microglia, have emerged as a critical focus for therapeutic strategies in inflammatory disorders. Morphological changes in mitochondria and production of reactive oxygen species also play important roles in exacerbating changes in metabolism.

Activation and Role of Astrocytes in Ischemic Stroke

Xin-Ya Shen et al.

Summary: Ischemic stroke leads to local brain tissue blood supply disorder, with astrocytes playing a crucial role in the response to the injury. Reactive astrocytes, activated after stroke, exhibit protective functions but also potentially harmful effects. Their spatial and temporal expression determines the diverse functions of astrocytes in the context of ischemic stroke's complex cascade reactions.

Encouraging an excitable brain state: mechanisms of brain repair in stroke

Mary T. Joy et al.

Summary: Stroke induces a plastic state in the brain, leading to the formation of new axons, synapses, and remapping of sensory-motor functions, facilitating motor recovery. The period of increased plasticity after stroke is driven by molecular systems related to learning and memory, resulting in changes in neuronal excitability. Understanding the mechanisms behind these changes is crucial for promoting functional recovery after stroke.
