Molecular Charge and Antibacterial Performance Relationships of Aggregation-Induced Emission Photosensitizers

Summary: Bacterial infections remain a major cause of morbidity worldwide due to drug resistance. Photodynamic therapy (PDT) offers a promising solution, but current photosensitizers (PSs) have broad-spectrum antibacterial effects and undesirable side effects. In this study, we synthesized aggregation-induced emission (AIE)-active PSs with varying charges and found that different charges influenced their antibacterial effects. Among the derivatives, TBTCP-SF lost its ability to bind and ablate bacteria, while TBTCP-QY and TBTCP-PEG showed enhanced binding and antibacterial activity. TBTCP-PEG significantly eliminated methicillin-resistant Staphylococcus aureus (MRSA) in vitro and in vivo. This work provides insights into using AIE-active PSs for targeted and efficient photodynamic ablation of bacteria.

Mechanically Robust Dissolving Microneedles Made of Supramolecular Photosensitizers for Effective Photodynamic Bacterial Biofilm Elimination

Summary: Antimicrobial photodynamic therapy (PDT) is a promising strategy for eradicating biofilms, but its efficacy is hindered by the low solubility and poor penetration of photosensitizers (PSs) into the biofilms. This study develops a dissolving microneedle (DMN) patch composed of a supramolecular PS for enhanced biofilm penetration and eradication. The DMN patch can efficiently penetrate the biofilms, allowing for optimal photodynamic elimination of bacterial biofilms. In addition, the DMN patch shows good biosafety in vivo for eradicating Staphylococcus aureus biofilm infection.

TPEQM-DMA can produce superoxide anion and hydroxyl radical through a low-O2-dependent type-I photochemical process, leading to disruption of cellular redox homeostasis, mitochondrial dysfunction, and increased peroxidized lipids, inducing cellular apoptosis and ferroptosis. TPEQM-DMA nanoparticles were prepared to improve its pharmacological properties and showed promising NIR-II fluorescence imaging-guided photodynamic therapy effect in vivo for tumors.

Raspberry-like gold nanozyme-hybrid liposomes for hypoxia-enhanced biofilm eradication

Visual Multifunctional Aggregation-Induced Emission-Based Bacterial Cellulose for Killing of Multidrug-Resistant Bacteria

Summary: To address the issue of multidrug-resistant (MDR) bacteria-related wound infections, researchers have developed a new antibacterial wound dressing that can prevent MDR bacteria infection and promote wound healing. The composite of aggregation-induced emission (AIE) molecule BITT and bacterial cellulose (BC) shows good transparency and allows for monitoring the wound healing process. The BC-BITT composites exhibit photodynamic and photothermal synergistic antibacterial effects under 660 nm laser irradiation and demonstrate excellent wound healing performance in a mouse full-thickness skin wound model infected by MDR bacteria, with negligible toxicity. This work provides a new approach for treating clinically troublesome wound infections.
