Rice RS2-9, which is bound by transcription factor OSH1, blocks enhancer-promoter interactions in plants

Huawei Liu et al.

Summary: Insulators in plants, like the RS2-9 insulator identified in rice, play a role in blocking enhancer-promoter interactions. The binding of O. sativa Homeobox 1 (OSH1) to RS2-9 is crucial for insulator function, suggesting a potential role for OSH1 in gene insulation and chromatin organization in plant genomes.

Genome-wide prediction of activating regulatory elements in rice by combining STARR-seq with FACS

Wei Tian et al.

Summary: In this study, the researchers found that the traditional STARR-seq method can be applied to rice protoplasts to identify enhancers, although with limited efficiency. They optimized the STARR-seq method by using fluorescence activating cell sorting (FACS) and minimizing PCR cycles and duplicates. The study revealed that AREs are associated with AT-rich regions and contain a motif recognized by the AP2/ERF family. AREs can be clustered into two groups corresponding to promoters and enhancers based on GC content preferences. Disruption of AREs resulted in abnormal gene expression. This work provides a promising method to identify AREs in plants.

CW198 acts as a genetic insulator to block enhancer-promoter interaction in plants

Li Jiang et al.

Summary: In this study, the researchers identified and validated an enhancer-blocking insulator called CW198 in plants. They found that this insulator can efficiently suppress the activation of enhancers in both Arabidopsis and tobacco plants. The properties of CW198 resemble those of the CTCF insulator in the mammalian genome.

The Dynamism of Transposon Methylation for Plant Development and Stress Adaptation

Muthusamy Ramakrishnan et al.

Summary: Plant development processes are regulated by epigenetic alterations, particularly by transposable elements (TEs), which interact with plant genomes and can cause genetic variability under stress conditions. TE-mediated epigenetic modifications lead to phenotypic diversity, genetic variation, and environmental stress tolerance in plants. High-throughput techniques have advanced the understanding of TE-mediated gene expression and suggest potential applications in crop breeding.

Atypical DNA methylation, sRNA-size distribution, and female gametogenesis in Utricularia gibba

Sergio Alan Cervantes-Perez et al.

Summary: This study reveals that compared to other angiosperms, Utricularia gibba has an unusual distribution of small RNAs and reduced global DNA methylation levels. Moreover, key genes involved in the RdDM pathway may lack compensatory paralogs or contain truncated proteins, potentially impacting the developmental processes in U. gibba.

Insulators in Plants: Progress and Open Questions

Amina Kurbidaeva et al.

Summary: Insulators, also known as boundary elements, play a key role in the formation and maintenance of topologically associated domains in higher eukaryotes. While extensively studied in fruit flies and mammals, knowledge about these elements in plant genomes is still limited. Current research focuses on identifying and determining the functions of plant insulators, laying the foundation for further studies in biotechnology.
