Disruption of the IL-33-ST2-AKT signaling axis impairs neurodevelopment by inhibiting microglial metabolic adaptation and phagocytic function

Danyang He et al.

Summary: This study reveals that cellular metabolism regulates microglial function during neurodevelopment. The IL-33 signaling axis, coupled with AKT activity, promotes mitochondrial metabolism and phagocytic function in microglia. This microglia-astrocyte circuit plays a crucial role in the metabolic adaptation and phagocytic function of microglia during early development.

Costimulation molecules differentially regulate the ERK-Zfp831 axis to shape T follicular helper cell differentiation

Siyuan Wan et al.

Summary: The study revealed that PMA + ionomycin stimulation combined with IL-6 can induce Tfh cell-like transcriptomic programs in vitro, and inhibition of ERK2 can enhance Tfh cell development. Additionally, ICOS lacks the ability to activate ERK, while Zfp831 promotes Tfh cell differentiation by upregulating the expression of Bcl6 and Tcf7.

Cytoplasmic DNA sensing by KU complex in aged CD4+ T cell potentiates T cell activation and aging-related autoimmune inflammation

Yan Wang et al.

Summary: The study reveals that the KU complex is abundantly expressed in CD4(+) T cells and enhances cell activation and proliferation in aged mice with EAE pathology by facilitating DNA-PKcs recruitment and phosphorylation of the kinase ZAK. A specific ZAK inhibitor can alleviate EAE pathology in aged mice.

Dynamic Dissection of the Endocytosis of Porcine Epidemic Diarrhea Coronavirus Cooperatively Mediated by Clathrin and Caveolae as Visualized by Single-Virus Tracking

Yangyang Li et al.

Summary: This study reveals a novel entry mechanism of porcine epidemic diarrhea coronavirus (PEDV) where clathrin and caveolae cooperate in mediating endocytosis. It also shows that about 20% of PEDVs undergo abortive endocytosis via clathrin-mediated endocytosis and caveolae-mediated endocytosis. Dynamics of viral entry, endosome trafficking, and fusion processes were dissected, providing new insights into coronavirus penetration of the cell membrane barrier.

Cinnamic acid inhibits Zika virus by inhibiting RdRp activity

Yuting Chen et al.

Summary: Recent study showed that cinnamic acid possesses antiviral properties against Zika virus (ZIKV) and may serve as a potential effective anti-ZIKV drug by inhibiting the post-entry stage of the replication cycle, reducing mortality of mice, and viral load in the blood.

Hypericin Inhibit Alpha-Coronavirus Replication by Targeting 3CL Protease

Yue Zhang et al.

Summary: The study demonstrated the antiviral effects of hypericin on PEDV and TGEV by targeting 3CLpro, suggesting its potential as a treatment for coronaviral infections. The binding pockets between hypericin and 3CLpro were highly conserved among alpha-CoVs.
