CD1d facilitates African swine fever virus entry into the host cells via clathrin-mediated endocytosis

Xin Chen et al.

Summary: African swine fever (ASF) is a highly contagious viral disease that has caused economic losses in the pig production industry. The mechanism of ASF virus (ASFV) entry into host cells is not fully understood. This study revealed that the host protein CD1d acts as a factor for ASFV entry, and its interaction with p72 facilitates viral infection. CD1d mediates viral entry through clathrin-mediated endocytosis and interacts with EPS15 to facilitate ASFV internalization. These findings highlight CD1d as a potential target for antiviral interventions.

Molecular mechanisms and therapeutic implications of tetrandrine and cepharanthine in T cell acute lymphoblastic leukemia and autoimmune diseases

Wencheng Xu et al.

Summary: Inappropriately activated T cells can lead to autoimmune diseases and T-ALL. Bisbenzylisoquinoline alkaloids tetrandrine and cepharanthine have shown potential in treating some autoimmune diseases and cancers, but further studies on their mechanisms of action and combined applications with other drugs are needed. These alkaloids influence the growth of activated T cells through various signaling pathways, and their P-glycoprotein inhibitory effect may be beneficial for refractory T cell-related diseases. Combined with glucocorticoids or anti-leukemia drugs, tetrandrine and cepharanthine offer a new therapeutic option for patients with drug-resistant conditions.

Antiviral drugs targeting endosomal membrane proteins inhibit distant animal and human pathogenic viruses

I Galindo et al.

Summary: The study demonstrates that drug compounds targeting endosomal membrane proteins show potent inhibitory effects on the entry of SARS-CoV-2, EBOV, and ASFV into cells, suggesting the potential of these compounds as broad-spectrum antivirals targeting the endocytic pathway proteins.

Identification of Novel Antiviral Compounds Targeting Entry of Hantaviruses

Jennifer Mayor et al.

Summary: Hemorrhagic fever viruses, including orthohantaviruses, arenaviruses, and filoviruses, cause severe human diseases and pose a significant challenge to public health. With limited therapeutic options and available vaccines, there is an urgent need for the development of novel antiviral agents. Inhibiting viral attachment and entry shows promise as a strategy for developing new treatments and preventing all subsequent steps in virus infection.

Macropinocytosis: Biology and mechanisms

Robert R. Kay

Summary: Macropinocytosis is a form of endocytosis performed by ruffles and cups of the plasma membrane, involving the engulfment of medium droplets into micron-sized vesicles for digestion and absorption. It is constitutive in certain immune cells and stimulated in others by growth factors, indicating a deep evolutionary history. The process is driven by the actin cytoskeleton and is increasingly studied for its medical implications in immune response, cancer development, and as a cellular entry point for viruses and drugs.

Tetrandrine suppresses the growth of human osteosarcoma cells by regulating multiple signaling pathways

Nan Wang et al.

Summary: Studies have shown that Tetrandrine (TET) can effectively prevent the proliferation, migration, and invasion of osteosarcoma cells, as well as promote apoptosis. The antitumor activity of TET may be mediated by its regulation of multiple signaling pathways, suggesting its potential as a treatment option for osteosarcoma.
