Extracellular vesicles, the emerging mirrors of brain physiopathology

Amanda Cano et al.

Summary: Extracellular vesicles are secreted by cells and play important roles in intercellular communication, immune responses, human reproduction, and synaptic plasticity. They have potential as biomarkers for diseases due to their biological relevance and easy accessibility. They can also be used as mirrors of brain diseases as they can cross the blood-brain barrier. This review explores their biogenesis, functions, and potential in clinical practice, as well as their role in major human brain diseases.

Production cell analysis and compound-based boosting of small extracellular vesicle secretion using a generic and scalable production platform

Christoph Keysberg et al.

Summary: This study presents a purification setup using tangential flow filtration (TFF) and fast-performance liquid chromatography (FPLC)-based size exclusion chromatography (SEC) suitable for industrial production of therapeutic extracellular vesicles (EVs). Potential host cell lines were evaluated, with telomerase-immortalized Wharton's jelly mesenchymal stromal cells (WJ-MSC/TERT273) found to secrete high amounts of regenerative EVs, and Cevec's amniocyte producer cells (R) (CAP (R)) and human embryonic kidney (HEK293) suspension cells suitable for designer EVs with high yields. Chemical adjuvants were used to enhance EV secretion in HEK293 cells, with the combination of fenoterol, iodoacetamide, and dinitrophenol increasing EV yield threefold and cellular secretion rate fivefold.

Extracellular vesicles facilitate large-scale dynamic exchange of proteins and RNA among cultured Chinese hamster ovary and human cells

Jessica Belliveau et al.

Summary: This study reveals a previously underestimated massive exchange of proteins and RNAs between cells mediated by extracellular vesicles (EVs), and estimates the per cell-specific rates of EV production in different cell types. This phenomenon of cell-to-cell material exchange through EVs may have implications for cell proliferation and cellular state in normal and tumor tissues.

Heterogeneity of mesenchymal stem cell-derived extracellular vesicles is highly impacted by the tissue/cell source and culture conditions

Ciarra Almeria et al.

Summary: Extracellular vesicles (EVs) are cell-derived membrane structures that play a major role in physiological and pathological processes by delivering biomolecules to target cells. Many effects previously attributed to cell-based therapies are actually mediated by EVs, suggesting the use of purified EVs instead of living cells for therapy. This review focuses on the heterogeneity of EVs derived from mesenchymal stem/stromal cells (MSC) and summarizes the upstream parameters that critically affect their therapeutic properties and biological functions.

Scalable Production of Extracellular Vesicles and Its Therapeutic Values: A Review

Chiew Yong Ng et al.

Summary: Extracellular vesicles (EVs) are small vesicles secreted by cells for intercellular communication. They are rich in biological components such as nucleic acids, lipids, and proteins, and play essential roles in tissue regeneration and disease modification. Researchers have employed various strategies, such as bioreactors, mechanical, electrical, thermal, and magnetic field stimulation, to improve the secretion of EVs by cultured cells. Additionally, techniques like nitrogen cavitation, porous membrane extrusion, and sonication have been used to prepare EV-mimetic nanovesicles. These interventions not only induce EV production but also modify their cargo and enhance their therapeutic potential.

Summary: Endosomal sorting complex required for transport (ESCRT) proteins assemble on membranes and remodel them. ESCRT-III proteins, the downstream components, can polymerize into filaments and spirals upon binding to negatively charged membranes. High-speed atomic force microscopy (HS-AFM) has provided detailed insights into the structure and dynamics of ESCRT-III. It has been used to study the four sequential steps in the ESCRT-III lifecycle: polymerization, morphology, dynamics, and depolymerization.

Small extracellular vesicles as a multicomponent biomarker platform in urinary tract carcinomas

K. Szeliski et al.

Summary: Extracellular vesicles are a diverse group of nano-sized vesicles released by all cells, containing various cargo and serving as a rich source of biomarkers for cancer diagnosis. Small extracellular vesicles, mainly composed of exosomes, have attracted significant attention in cancer diagnostic applications. Studies have evaluated the diagnostic potential of these vesicles in urinary tract carcinomas, identifying various biomarkers. sEVs, particularly non-coding RNAs, play an essential role in the cancer process and offer diagnostic and prognostic applications for UTC. The ease of obtaining biofluids rich in sEVs confirms their significance in UTC detection using liquid biopsy.
