Phospholipase D3 degrades mitochondrial DNA to regulate nucleotide signaling and APP metabolism

Summary: Phospholipase D3 polymorphisms are associated with late-onset Alzheimer's disease (LOAD), but the underlying mechanisms are unclear. This study reveals that Phospholipase D3 processes mitochondrial DNA in lysosomes, leading to lysosomal dysfunction and abnormal degradation of amyloid precursor protein. The accumulation of mitochondrial DNA in lysosomes activates cGAS-STING signaling, resulting in dysregulation of autophagy and cholesterol metabolism, which contributes to the pathogenesis of LOAD.

Origin of iodine preferential attack at sulfur in phosphorothioate and subsequent P-O or P-S bond dissociation

Summary: Iodine-induced cleavage at phosphorothioate DNA has potential applications in detecting and sequencing bacterial DNA. However, the cleavage mechanism and specific selectivity at the modified site are not well established. This study reveals that iodine selectively binds to the sulfur atom of the phosphorothioate link and triggers P-O bond cleavage through phosphotriester-like hydrolysis. The cleavage process also results in unwanted P-S/P-O conversions and loss of the phosphorothioate handle.

Cysteine Oxidation in Proteins: Structure, Biophysics, and Simulation

Cleavage of DNA and RNA by PLD3 and PLD4 limits autoinflammatory triggering by multiple sensors

This suggests that both endosomal TLR and cytoplasmic/STING nucleic acid sensing pathways are regulated by PLD3 and PLD4, offering potential implications for treating nucleic acid-driven inflammatory diseases.

Quantification and characterization of the 5 ' exonuclease activity of the lysosomal nuclease PLD3 by a novel cell-based assay

Summary: Phospholipase D3 (PLD3) and phospholipase D4 (PLD4) are recently described lysosomal nucleases associated with various diseases. A cell-based assay was developed to quantify the 5' exonuclease activity of PLD3 and PLD4, revealing PLD3 as the primary acid 5' exonuclease with higher specific activity in HeLa cells. Experiments on genetic variants of PLD3 found that some variants result in reduced activity.
