A randomized clinical trial of granulocyte macrophage colony stimulating factor mouthwash for oral mucositis in head and neck cancer

Summary: This study aimed to compare the effectiveness of granulocyte macrophage colony stimulating factor (GM-CSF) mouthwash and Salt and Soda mouthwash in the prevention and treatment of radiation therapy-induced oral mucositis (OM). The results showed no significant differences between GM-CSF and Salt and Soda mouthwash in the occurrence and grades of OM during the prevention phase, as well as in functional status, pain, chewing, and swallowing during and after radiation therapy. Therefore, Salt and Soda mouthwash can be used instead of GM-CSF mouthwash to save costs.

IL-3 signalling in the tumour microenvironment shapes the immune response via tumour endothelial cell-derived extracellular vesicles

Summary: This study investigated the clinical impact of targeting the IL-3 signaling in the microenvironment of solid tumors and the modulation of PD-L1 expression in tumor cells and peripheral blood mononuclear cells (PBMC) through IL-3Ra blockade. The study found that IL-3 controls the expression of its receptor on tumor-derived endothelial cells (TEC) and PBMC and that IL-3 secretion increases PD-L1 expression in myeloid cells. Additionally, the study demonstrated that TEC-derived extracellular vesicles can reshape the anti-tumor immune response, possibly through the enrichment of miR-214.

Summary: This article reviews the occurrence and mechanisms of abnormal pathological hematopoiesis in solid tumors, with a focus on the hematopoietic dysfunction induced by tumor-released G-CSF and GM-CSF. These cytokines, known as facilitators of myelopoiesis and HSC mobilization, can also have either promoting or inhibitory effects on tumor growth depending on the tumor type. The study found that G-CSF and GM-CSF can significantly enhance hematopoiesis and induce abnormal lineage differentiation in multiple cancer types, with potential implications for tumor metastasis.

Enhanced neutrophil to lymphocyte ratio may contribute to increased mortality and decreased survival in breast cancer cases.
