Advances of nanoparticles as drug delivery systems for disease diagnosis and treatment

Today, there have been significant advancements in nanoparticles, including liposomes, solid lipid nanoparticles, inorganic nanoparticles, micelles, nanovaccines, and nano-antibodies. These nanoparticles have shown great potential in the treatment and diagnosis of challenging diseases such as cancer and neurodegenerative diseases, making them promising drug carriers and therapeutic agents in the future.

Polymer-based biomaterials for pharmaceutical and biomedical applications: A focus on topical drug administration

Synthesis and characterization of magnetic nanoparticles-grafted-hyaluronic acid/β-cyclodextrin as a novel pH-sensetive nanocarrier for targeted delivery of doxorubicin

Summary: pH-sensitive magnetic nanocarrier was developed for doxorubicin delivery by grafting hyaluronic acid/beta-cyclodextrin onto Fe3O4 magnetic nanoparticles. The nanocarrier demonstrated strong pH-dependent release behavior, with higher drug release at pH 5.6 compared to pH 7.4. The release mechanism was determined to be controlled by Fickian diffusion kinetic. The surface modification of the magnetic nanoparticles greatly influenced drug sorption and was more compatible with Langmuir isotherm and pseudo-second-order kinetic models. Thermodynamic parameters indicated that the doxorubicin adsorption was endothermic and spontaneous, suggesting physiochemical sorption. These findings highlight the potential of pH-sensitive nanocarriers for drug delivery.

pH-responsive magnetic Fe3O4/carboxymethyl chitosan/aminated lignosulfonate nanoparticles with uniform size for targeted drug loading

Summary: A magnetic delivery system for targeted drug was developed using aminated lignosulfonate (ALS) and carboxymethyl chitosan (CMCS) to fabricate nano Fe3O4 with pH responsiveness. The system successfully loaded doxorubicin hydrochloride (DOX) and demonstrated controlled release of the drug. The particle size ranged from 79.9-169.9 nm under optimized conditions and exhibited excellent pH responsiveness. The drug-loaded particles showed significant growth inhibition on cancer cells.

Polysaccharide-based nanocarriers for efficient transvascular drug delivery

Summary: Polysaccharide-based nanocarriers (PBNs) have gained significant attention due to their biocompatibility, cost-effectiveness, availability, and versatility in loading anticancer agents. However, the heterogeneous tumor structures compromise the enhanced permeability and retention (EPR) effect, hindering efficient drug delivery. Overcoming the biological barriers for extravasation from tumor vessels is crucial for optimizing PBNs. Various strategies, including nanocarrier property regulation and tumor structure remodeling, have been developed to enhance the EPR effect. This review summarizes recent advances in the design of PBNs with enhanced transvascular transport and discusses the challenges in understanding the transendothelial mechanism of PBNs and their interactions with other drug delivery steps.

Cladosporium protease/doxorubicin decorated Fe3O4@SiO2 nanocomposite: An efficient nanoparticle for drug delivery and combating breast cancer

Summary: Fe3O4/SiO2 nanocomposite was synthesized and decorated with an extracellular Cladosporium protease, doxorubicin, plus C. protease/DOX molecules. The C. Protease was purified using the mAzo method and its molecular weight was determined as 70 kDa by SDS-PAGE. The nanocomposite was characterized using various techniques. The cytotoxic effect of the decorated nanocomposites on the MCF-7 cancer cell line was studied, showing a 7.5% cell viability.

Functionalisation of Graphene Sensor Surfaces for the Specific Detection of Biomarkers

Summary: We present a controllable and specific functionalisation approach for graphene field-effect transistors (GFETs) in order to detect small physiologically active molecules. The key step involves the noncovalent functionalisation of graphene with perylene bisimide (PBI) molecules directly on the growth substrate. This method allows for the homogeneous self-assembly of PBI molecules on graphene, followed by the immobilization of antibodies. The functionalised GFET devices showed excellent performance in terms of atomic force microscopy, Raman spectroscopy, and electrical measurements compared to conventional functionalisation methods. Specific sensing of small molecules such as methamphetamine and cortisol was achieved by monitoring the electrical changes of the functionalised GFET devices. The sensors were able to detect methamphetamine down to a concentration of 300 ng ml(-1).

Comparative study of the efficiency of silicon carbide, boron nitride and carbon nanotube to deliver cancerous drug, azacitidine: A DFT study

Summary: In this study, we compared the efficiency of three different nanotubes (carbon nanotube, boron nitride nanotube, and silicon carbide nanotube) in delivering the cancer drug Azacitidine (AZD). Parameters such as adsorption energy, electrostatic potential map, and reduced density gradient were analyzed to understand the encapsulation process of AZD in these nanotubes. The results showed that BNNT and SiCNT had a higher adsorption energy and stronger binding affinity with AZD compared to CNT. Thermodynamic parameters also revealed that the encapsulation process was spontaneous and exothermic, especially with BNNT and SiCNT. Molecular dynamics simulation supported the encapsulation process of AZD in the nanotubes at room temperature. These findings suggest the potential of these nanotubes as drug delivery systems in nanomedicine.

Magnetic nanoparticle embedded chitosan-based polymeric network for the hydrophobic drug delivery of paclitaxel

Summary: Safe delivery of hydrophobic drugs to the tumor site is a challenging task for the scientific community. In this study, iron oxide nanoparticles coated chitosan with [2-(methacryloyloxy) ethyl] trimethyl ammonium chloride (METAC) (CS-IONPs-METAC-PTX) were developed as a drug carrier for hydrophobic drug delivery. The drug carrier exhibited excellent therapeutic efficacy and selectivity when tested in cell lines, demonstrating its potential for PTX delivery.

Co-delivery of microRNA-150 and quercetin by lipid nanoparticles (LNPs) for the targeted treatment of age-related macular degeneration (AMD)

Summary: Age-related macular degeneration (AMD) is a disease that causes severe vision loss in middle-aged and elderly patients due to choroidal neovascularization (CNV). Current treatments for CNV are ineffective and can have adverse effects. This study developed solid lipid nanoparticles modified with an Asp-Gly-Arg peptide to target endothelial cells during abnormal angiogenesis. These nanoparticles successfully co-delivered microRNA-150 (mR150) and the antiangiogenic drug quercetin, inhibiting CNV without retinal toxicity in a mouse model.

Mesoporous silica nanoparticles: Synthesis and multifaceted functionalization for controlled drug delivery

Summary: Inorganic mesoporous silica nanoparticles (MSN) have been extensively studied as porous materials for drug delivery due to their easy synthesis methods, tunable pore size, biocompatibility, high loading efficiency, and surface functionalization properties. This review discusses various synthesis approaches and functionalization strategies of MSN for active and passive targeting, including controlled drug delivery with stimuli-sensitive gatekeepers. Additionally, the review explores stimuli-responsive drug delivery approaches targeting pH, redox, enzyme, temperature, light, etc. MSN-based nanoparticles have also been used to achieve enhanced on-demand release and reduce non-specific drug leakage through dual and triple stimuli. The versatility of MSNs makes them promising drug delivery vectors.

Controlling the Binding Efficiency of Surface Confined Antibodies through the Design of Mixed Self-Assembled Monolayers

Summary: A variety of electronic and optoelectronic devices have been developed recently for biosensing applications, with a focus on a detecting interface that incorporates biorecognition elements anchored by self-assembled monolayers (SAMs) on a gold surface. This study investigates the effect of different SAM structures on the packing efficiency of antibodies and the analytical performance of sensors by performing surface plasmon resonance (SPR) characterization of anti-p24 anchored on these SAMs. The results show that the choice of SAM significantly affects the binding efficiency of antibodies against the HIV p24 protein, and density functional theory calculations provide a molecular understanding of these observations.

Drug content on anticancer efficacy of self-assembling ketal-linked dextran-paclitaxel conjugates

Depot unilamellar liposomes to sustain transscleral drug Co-delivery for ophthalmic infection therapy

Summary: Periocular routes have great potential for the treatment of chronic eye diseases by exploiting the scleral structure. In this study, phosphatidylcholine-based liposomes (LIP-DM) were developed to deliver water-soluble steroid dexamethasone and antibiotics moxifloxacin through the sclera. The results demonstrated that LIP-DM exhibited differential release of the two drugs, with fast release of moxifloxacin and slow release of dexamethasone, leading to increased transscleral contact time and enhanced antibacterial and anti-inflammatory effects.

Summary: Lipid nanoparticles containing ionizable aminolipids play a crucial role in delivering nucleic-acid-based therapeutics. This study investigated the distribution and effects of Lipid-5, an ionizable aminolipid, on lipid bilayers using molecular dynamics simulations. The results showed that high concentrations of ionizable aminolipids led to the formation of a disordered aggregate of neutral aminolipids in the membrane interior, preferentially incorporating cholesterol. Magnetic graphene oxide nanocomposite as dual-mode genosensor for ultrasensitive detection of oncogenic microRNA

Summary: This study designed multifunctional nanosheets for the capture, detection, and quantification of miRNA 183-5p from prostate cancer cells using a disposable printed electrode and a portable potentiostat. Magnetic reduced graphene oxide (MrGO) was used as the starting nanocomposite to analyze miRNA. Two strategies were used for miRNA detection, and the intercalator method showed better performance. The detection limits of miRNA in human serum and urine were 3.73 and 0.86 aM, with a linear range from 0.1 nM to over 1 aM. The incorporation of MrGO, screen-printed carbon electrodes, and portable potentiostat simplified the analysis process and reduced time and cost.

Highly sensitive and selective fluorescence sensor based on zirconia nanotube array films for Cu2+detection

Summary: ZrO2 nanotube array films (ZNA) were prepared through anodisation method and modified with gamma-aminopropyltriethoxysilane (APTES) to obtain amino-functionalized ZNA (ZNA-NH2). Cu2+ ion detection was achieved by immobilizing Cu2+ ions and RB on ZNA and ZNA-NH2, and measuring the change in fluorescence intensity of RB. The effects of APTES and RB concentrations on the detection performances were studied, and ZNA-NH2 exhibited significantly better fluorescence detection performances compared to ZNA. The optimal detection performances for Cu2+ ions were obtained with an RB concentration of 10 mg/L and an APTES concentration of 0.4 M, with a minimum detection limit of 0.68 nM (S/N = 3) and a linear range from 1 x 10-3 to 3 x 102 μM under the optimal experimental conditions. The carriers could be regenerated and reused after being soaked in acetic acid solution. This work can be applied to the detection of Cu2+ ions in real seawater.

Aptamer-conjugated gold nanoparticles for targeted paclitaxel delivery and photothermal therapy in breast cancer

Cucurbit[n]uril-Immobilized Sensor Arrays for Indicator-Displacement Assays of Small Bioactive Metabolites

Summary: The patterned immobilization of cucurbit[n]urils (CBn) into multiplexed microarrays enhances sensitivity and allows for the detection of small molecule analytes through indicator-displacement assays. Integration of CBn/indicator microarrays into microfluidic channels provides efficient real-time monitoring of the sensing process and reduces the required analyte volume. The concept is further extended to differential sensing of analytes on diplex or multiplex CBn/indicator microarrays, offering a route for multicomponent sensing of small molecule analytes in complex liquids.
