Summary: High concentrations of electrophilic lipid alkenals have cytotoxicity and disease implications, while low concentrations induce antioxidative stress responses. The clearance pathway of alkenals involves conjugation to glutathione (GSH) via Michael addition catalyzed by GSTA4-4. We propose that GSTA4-4 contributes to lipid metabolism homeostasis based on its catalytic competence in retro-Michael addition with NE and the antioxidant function of low alkenal concentrations.

Oxidative distress in aging and age-related diseases: Spatiotemporal dysregulation of protein oxidation and degradation

Summary: The concept of oxidative distress arises from the imbalance between oxidants and antioxidants, leading to biomolecular damage. Compromised spatiotemporal control of reactive species generation and removal underlies protein damage and dysfunction of protein degradation machinery, resulting in aging and age-related diseases. The 20S proteasome and autophagy-lysosome pathways play crucial roles in the degradation and elimination of oxidatively damaged proteins. Future investigations in this field may lead to the discovery of new drugs targeting dysregulated cell signaling and protein degradation machinery to combat aging and age-related chronic diseases.

Insight into the Impact of Oxidative Stress on the Barrier Properties of Lipid Bilayer Models

Summary: Cold physical plasma, as a new field of oxidative stress-based therapy, has the potential to generate a variety of reactive oxygen and nitrogen species (RONS), making it a promising tool for biomedical applications. However, the impact of plasma-derived RONS on cell membrane lipids and properties is not fully understood. This study investigated the changes in lipid bilayer functionality and revealed the strong impact of cholesterol on membrane permeabilization by plasma-derived species. Additionally, the chemical composition of the head group was found to influence the maintenance of barrier properties, with phosphatidylcholines being more susceptible than other lipid classes.

L-Arginine prevents 4-hydroxy-2-nonenal accumulation and depresses inflammation via inhibiting NF-κB activation

Summary: This study explored the relationship between the inhibition of HNE accumulation and the depression of inflammation by examining the effects of L-arginine intake in growing rats. The results showed that L-arginine reduced hepatic accumulation of HNE and suppressed inflammation in rats. This anti-inflammatory effect was attributed to the upregulation of IL-10 and the downregulation of pro-inflammatory cytokines, as well as the inhibition of NF-kappa B activation via the downregulation of PI3K/Akt.

Determination of acrolein generation pathways from linoleic acid and linolenic acid: increment by photo irradiation

Summary: In this study, the authors investigated the pathway of acrolein formation from type II photo-oxidation in edible oils. They found that linoleic acid (LA) and linolenic acid (LnA) are significant sources of acrolein formation through singlet oxygen oxidation. The amount of acrolein formed from oils high in LA increased with photo irradiation. This information can be used to guide oil storage and processing conditions to reduce exposure and dietary intake of acrolein.

In vitro dosimetry analyses for acrolein exposure in normal human lung epithelial cells and human lung cancer cells
