The E3 Ubiquitin Ligase RNF5 Facilitates SARS-CoV-2 Membrane Protein-Mediated Virion Release

Zhen Yuan et al.

Summary: In this study, the role of the E3 ligase RNF5 and the deubiquitinating enzyme POH1 in the release of SARS-CoV-2 was investigated. It was found that RNF5 mediates the ubiquitination of SARS-CoV-2 M, enhancing the interaction with the viral envelope protein E, while POH1 negatively regulates this process. The M-E complex plays a crucial role in viral maturation and release, and M uses autophagosomes for virion release, dependent on RNF5-mediated ubiquitin modification and M-E interaction.

Interaction of Nucleolin with the Fusion Protein of Avian Metapneumovirus Subgroup C Contributes to Viral Replication

Dedong Wang et al.

Summary: It has been found that avian metapneumovirus subgroup C (aMPV/C) infection can cause changes in the subcellular localization of nucleolin (NCL) and NCL plays a role in aMPV/C replication. Knocking down NCL using siRNA or CRISPR/Cas9 significantly reduces aMPV/C replication, while overexpression of NCL increases viral replication. The interaction between the aMPV/C fusion (F) protein and NCL is important for viral replication, and AS1411 disrupts this interaction, inhibiting aMPV/C replication.

E3 ligase Nedd4l promotes antiviral innate immunity by catalyzing K29-linked cysteine ubiquitination of TRAF3

Peng Gao et al.

Summary: The study highlights the crucial role of Nedd4l in regulating antiviral immunity through promoting ubiquitination of TRAF3, and disruption of Nedd4l impairs the antiviral immune response both in vitro and in vivo.

The PB1 protein of influenza A virus inhibits the innate immune response by targeting MAVS for NBR1-mediated selective autophagic degradation

Yan Zeng et al.

Summary: The PB1 protein of H7N9 influenza virus was identified as a novel negative regulator that enhances the degradation of MAVS, inhibiting the innate immune antiviral response and facilitating virus replication. This PB1-RNF5-MAVS-NBR1 axis provides insights into flu viruses' evasion strategy involving selective autophagy and innate signaling pathways.

Nucleolin Promotes IRES-Driven Translation of Foot-and-Mouth Disease Virus by Supporting the Assembly of Translation Initiation Complexes

Shichong Han et al.

Summary: Nucleolin (NCL) is identified as a protein interacting with foot-and-mouth disease virus (FMDV) IRES and regulating translation of various IRES-containing viruses. NCL plays a substantial role in viral replication by directly regulating viral protein production and indirectly influencing FMDV RNA synthesis. The cytoplasmic relocalization of NCL during FMDV infection is crucial for viral IRES-driven translation, highlighting its importance as a potential target for antiviral interventions.

Overexpression of ARHGAP30 suppresses growth of cervical cancer cells by downregulating ribosome biogenesis

Aijia Wu et al.

Summary: The study identified ARHGAP30 as the only downregulated RhoGAP in cervical cancer, associated with shorter survival of patients. Overexpression of ARHGAP30 inhibited cell proliferation and migration, while promoting ubiquitination of NCL to suppress ribosome biogenesis and protein synthesis in cervical cancer cells. ARHGAP30's effects on ribosome biogenesis and proliferation were independent of its RhoGAP activity.
