Tumor-associated fibrosis impairs the response to immunotherapy

Angha Naik et al.

Summary: Previously, impaired responses to immunotherapy in cancer had been attributed mainly to inherent tumor characteristics, but mapping responses in clinical trials revealed a commonality: tumors with a rich fibrotic stroma respond poorly. This prompted a closer look at tumor cell extrinsic factors such as the surrounding tumor microenvironment and the role of cancer-associated fibrosis in immunotherapy failure. Recent studies have established a complex interconnection between fibrosis and treatment efficacy.

CXCL1: Gene, Promoter, Regulation of Expression, mRNA Stability, Regulation of Activity in the Intercellular Space

Jan Korbecki et al.

Summary: CXCL1 is an important chemokine involved in the development of inflammatory diseases. Its expression is elevated in inflammatory reactions and it plays important roles in physiology. This paper presents the mechanisms of CXCL1 expression regulation with a focus on cancer, including transcription regulation, mRNA stability, and activity regulation in the extracellular space. The role of CXCL1 in cancer and potential therapeutic approaches are also discussed.

Chemokines as Regulators of Neutrophils: Focus on Tumors, Therapeutic Targeting, and Immunotherapy

Raffaella Bonecchi et al.

Summary: Neutrophils, the main leukocyte subset in human blood, have a crucial role in defending against infections and also play a significant role in the tumor stroma. Tumor-associated neutrophils can have both protumor and antitumor activities, and chemokines and chemokine receptors are considered targets to improve their antitumoral function in cancer immunotherapy. Targeting chemokines or chemokine receptors can modulate neutrophil activity and enhance their antitumor properties.

CXCR2 Is Essential for Radiation-Induced Intestinal Injury by Initiating Neutrophil Infiltration

Jing Hu et al.

Summary: Neutrophils play a critical role in radiation-induced intestinal injury, with migration mediated by the CXCR2 receptor. Inhibiting the CXCR2 signaling pathway effectively suppresses neutrophil chemotaxis and tissue damage.

Radiation-induced IL-1β expression and secretion promote cancer cell migration/invasion via activation of the NF-κB-RIP1 pathway

A-Ram Kang et al.

Summary: The study demonstrates the role of IL-1 beta in promoting migration/invasion in A549 lung cancer cells induced by gamma-ionizing radiation (IR), through RIP1 upregulation. IR-induced IL-1 beta is found downstream of NF-kappa B-RIP1 signaling pathway, and an autocrine loop between IL-1 beta and IL-1 RI/II may contribute to IR-induced EMT and migration/invasion, ultimately promoting metastasis.

Increasing neutrophil-to-lymphocyte ratio following radiation is a poor prognostic factor and directly correlates with splenic radiation dose in pancreatic cancer

Adam R. Wolfe et al.

Summary: The study found that an elevated ANLR after radiation therapy significantly predicted worse overall survival (OS) and decreased odds of achieving surgical resection. Furthermore, ANLR was correlated with higher splenic doses, suggesting the spleen may be an important organ at risk.

The AIM2 and NLRP3 inflammasomes trigger IL-1-mediated antitumor effects during radiation

Chuanhui Han et al.

Summary: Research showed that tumors lacking certain molecules in the inflammasome pathway were sensitive to radiation treatment in mice, while dual deficiency in multiple molecules resulted in radioresistance. The study also demonstrated that activation of AIM2 and NLRP3 inflammasomes by irradiated tumors played a role in inducing antitumor effects through IL-1 signaling in dendritic cells.
