Spike and nsp6 are key determinants of SARS-CoV-2 Omicron BA.1 attenuation

Da-Yuan Chen et al.

Summary: The Omicron variant of SARS-CoV-2 is highly immune evasive but less virulent compared to other major viral variants. It has a large number of mutations in the spike protein, which is the main driver of these traits. The virus bearing Omicron S can efficiently escape vaccine-induced immunity, replicate in cells, and cause less severe disease, but its pathogenicity is determined by mutations both in and outside of the spike protein.

Rapid assembly of SARS-CoV-2 genomes reveals attenuation of the Omicron BA.1 variant through NSP6

Taha Y. Taha et al.

Summary: Researchers developed a plasmid-based strategy for assembling and rescuing viral genomes, which allows efficient construction of replication-competent viruses. Using this strategy, they discovered that mutations in the nonstructural protein 6 of the Omicron variant contribute to viral attenuation.

SARS-CoV-2 nonstructural protein 6 from Alpha to Omicron: evolution of a transmembrane protein

Shuchen Feng et al.

Summary: We recently reported a surprising role for nsp6 replicase component, in addition to spike changes, in the attenuation of Omicron BA.1. We documented a triple-amino-acid deletion in a predicted lumenal domain of nsp6 found in multiple variants of SARS-CoV-2, including all recent Omicron lineages. Modeling the predicted structure of nsp6 revealed a conserved multipass transmembrane architecture in the Coronaviridae family.

Unresolved questions in the zoonotic transmission of MERS

Malik Peiris et al.

Summary: The Middle East Respiratory Syndrome-coronavirus (MERS-CoV) primarily spreads to humans through dromedary camels in Africa, the Middle East, and Central or South Asia, with limited human-to-human transmission. Only cases in humans from the Arabian Peninsula have been reported so far, and no zoonotic MERS cases have been found in Africa or Asia, despite the high density of infected camels. Further investigation into this disparity is crucial due to the pandemic potential of MERS-CoV.

The role of NSP6 in the biogenesis of the SARS-CoV-2 replication organelle

Simona Ricciardi et al.

Summary: The replication organelle of SARS-CoV-2 is generated by viral non-structural proteins (NSPs) NSP3 and NSP4, while NSP6 plays a role in connecting ER membranes and organizing DMV clusters. NSP6 also mediates the contact between SARS-CoV-2 and lipid droplets (LDs), providing a means to refurbish the replication organelle with LD-derived lipids.

Ubiquitination of SARS-CoV-2 NSP6 and ORF7a Facilitates NF-kappa B Activation

Hironori Nishitsuji et al.

Summary: This study reveals the molecular mechanisms behind the induction of proinflammatory cytokines by the SARS-CoV-2 virus. The NSP6 and ORF7a proteins activate the NF-kappa B pathway through interactions with TAK1. The polyubiquitination of NSP6 and ORF7a is essential for NF-kappa B activation, with TRIM13 and RNF121 playing key roles in this process. These findings provide novel insights into the pathogenesis of SARS-CoV-2 and the host immune response.

Evaluating the Effects of SARS-CoV-2 Spike Mutation D614G on Transmissibility and Pathogenicity

Erik Volz et al.

Summary: The study suggests a positive selection for the SARS-CoV-2 spike protein variant D614G in the UK, but no evidence of differences in COVID-19 mortality or clinical severity in patients infected with this variant. 614G is associated with higher viral load and younger age of patients compared to 614D.
