Signaling Pathways of the Insulin-like Growth Factor Binding Proteins

Robert C. Baxter

Summary: The 6 high-affinity insulin-like growth factor binding proteins (IGFBPs) have diverse functions in cell signaling and can impact cell survival, migration, metabolism, and other cellular functions. They interact with multiple pathways and regulatory proteins, including integrins and transforming growth factor beta family receptors. IGFBPs can also regulate nuclear hormone receptors, senescence, DNA damage repair, and immune functions. Understanding the complexity of IGFBPs' interactions may provide therapeutic opportunities for modulating IGFBP-dependent signaling.

Bipotent transitional liver progenitor cells contribute to liver regeneration

Wenjuan Pu et al.

Summary: Transitional liver progenitor cells (TLPCs), derived from biliary epithelial cells (BECs), differentiate into hepatocytes after severe liver damage. Notch and WNT/β-catenin signaling regulate the conversions of BEC-to-TLPC and TLPC-to-hepatocyte, respectively.

Premature senescence of the liver in Alagille patients

Giulia Jannone et al.

Summary: This study investigated premature senescence and senescence-associated secretory phenotype (SASP) in ALGS livers. The results showed advanced premature senescence in the livers of ALGS patients, despite Jagged1 mutation. This study highlights the complexity of senescence and SASP development pathways.

Intrahepatic cholangiocyte regeneration from an Fgf-dependent extrahepatic progenitor niche in a zebrafish model of Alagille Syndrome

Chengjian Zhao et al.

Summary: The study demonstrates that resumption of Notch signaling in zebrafish can lead to regeneration of intrahepatic duct (IHD) cells from multipotent progenitors in an Fgf-dependent extrahepatic stem cell niche. This finding suggests that enhancing Jagged/Notch signaling may promote regeneration of IHD cells in patients with Alagille Syndrome.

Two distinct Notch signals, Delta-like 4/Notch1 and Jagged-1/Notch2, antagonistically regulate chemical hepatocarcinogenesis in mice

Summary: The Dll4/Notch1 signal promotes the progression of hepatocellular carcinoma (HCC), while the Jag1/Notch2 signal antagonistically suppresses it. This study reveals the opposing effects of two distinct Notch signals on HCC development.

Sex differences and risk factors for bleeding in Alagille syndrome

Simona Hankeova et al.

Summary: This study found significant sex differences in spontaneous bleeding events reported by ALGS patients, with more girls than boys experiencing intracranial hemorrhage. Jag1(Ndr/Ndr) mice displayed various vascular defects, and retinography proved to be a non-invasive method to analyze vascular changes in patients.

Summary: Organoid culture systems derived from pluripotent and adult hepatobiliary cells are used to model organ structure and function. These systems have been applied to study various liver and biliary diseases, as well as for drug testing and cancer sample biobanking. Challenges include cellular immaturity, imperfect tissue structure modeling, and lack of standardized protocols.

Simona Hankeova et al.

Summary: The technique described in the study, double resin casting micro-computed tomography (DUCT), involves injecting two different resins into the portal vein and bile duct, followed by tissue fixation, to visualize the three-dimensional structure of the liver. DUCT can be applied to postnatal and adult mouse liver and other tubular networks.
