Environmental and genetic associations with aberrant early-life gut microbial maturation in childhood asthma

Summary: This study aimed to determine the roles of environmental exposures and 17q12-21 locus genotype in the maturation of the early-life microbiome in childhood asthma. The findings revealed that accelerated and delayed fecal microbial maturation at different ages were associated with asthma. Fecal Bacteroides was reduced in association with subsequent asthma and lower microbial maturation. Breast-feeding and asthma association was mediated by microbial maturation at 3-6 months. The intestinal microbiome and 17q12-21 genotype exhibited additive and independent effects on childhood asthma risk.

Characteristics and Mechanisms of a Sphingolipid-associated Childhood Asthma Endotype

Summary: This study explored a childhood asthma endotype characterized by reduced sphingolipid concentrations, early onset, and increased airway resistance. Lower concentrations of ceramides and sphingomyelins at 6 months correlated with an increased asthma risk before age 3, while reduced levels of key phosphosphingolipids at age 6 were associated with higher airway resistance. These associations were influenced by the 17q21 genotype and nasal gene expression of SPT.

Summary: Prenatal vitamin D-3 supplementation has a protective effect against offspring asthma/recurring wheeze, which is influenced by maternal 17q21 genotype variation. Offspring of mothers with low-risk genotype show significant reduction in risk with high-dose vitamin D-3 supplementation, while no difference was observed in offspring of mothers with high-risk genotype.
