Exchangeable HaloTag Ligands for Super-Resolution Fluorescence Microscopy

Summary: The specific and covalent labeling of the protein HaloTag with fluorescent probes in living cells makes it a powerful tool for bioimaging. However, the irreversible attachment of the probe to HaloTag precludes imaging applications that require transient binding of the probe and comes with the risk of irreversible photobleaching. Here, we introduce exchangeable ligands for fluorescence labeling of HaloTag (xHTLs) that reversibly bind to HaloTag and that can be coupled to rhodamines of different colors.

Phosphorescent iridium(III) complexes as lifetime-based biological sensors for photoluminescence lifetime imaging microscopy

Summary: Luminescent iridium(III) complexes have been extensively studied as intracellular sensors for photoluminescence microscopy imaging due to their high phosphorescence quantum yields, photostability, good cell membrane permeability, and rich and sensitive photophysical properties. Photoluminescence lifetime imaging microscopy (PLIM) allows for visualizing and mapping the decay of luminescent dyes in the time domain during cellular imaging. This review article focuses on the design and applications of lifetime-responsive phosphorescent iridium(III) complexes as imaging reagents for mapping environmental parameters and sensing analytes of interest in intracellular conditions.

Luminescence Lifetime Imaging Based on Lanthanide Nanoparticles

Summary: Luminescence lifetime imaging has advantages over traditional optical imaging in terms of precise quantification detection, and the long luminescence lifetime of lanthanide-doped nanoparticles (LnNPs) has attracted great attention. LnNPs with long luminescence lifetime not only make it easier to filter out background signals during imaging, but also provide a wide range of tunable lifetime. This article introduces the luminescence mechanisms of LnNPs, strategies for luminescence lifetime modulation, data acquisition methods, and cutting-edge applications in lifetime imaging systems, as well as prospects for improving technologies and extending applications of luminescence lifetime imaging.

Photostability of luminescent europium(III) hexafluoroacetylacetonates: Combined theoretical and experimental study

Summary: The photostability and molecular structure of four europium hexafluoroacetylacetonates were studied in this work, with a focus on the unique behavior of the Hfaa ligand during photoexcitation and its significant impact on the photostability of the compounds. Results showed that the presence of the TPPO ligand in the complex contributed to increased photostability by decreasing the antibonding character of the HOMO during photoexcitation.
