A Bright Two-Photon Lipid Droplets Probe with Viscosity-Enhanced Solvatochromic Emission for Visualizing Lipid Metabolic Disorders in Deep Tissues

Summary: The polarity of lipid droplets (LDs) is important in pathological processes associated with abnormal lipid metabolism. Monitoring LDs polarity in cells and tissues is crucial in biomedical research and clinical diagnosis. However, developing suitable probes for deep tissue imaging of LDs is challenging. In this study, a push-pull fluorescent luminogen with aggregation-induced emission and solvatochromism is developed, which successfully stains LDs in various mouse tissues and maps their polarity distribution. The excellent properties of this probe make it a promising candidate for investigating LDs-associated physiological and pathological processes in live biological samples.

pH-Triggered Charge Reversible Fluorescent Probe for Simultaneous Imaging of Lipid Droplets and Nucleoli in Living Cells

Summary: Cooperation between organelles, such as lipid droplets (LDs) and nucleoli, is crucial for maintaining normal cellular functions. However, there has been limited in situ observation of the interaction between them due to the lack of suitable tools. In this study, a pH-triggered charge reversible fluorescent probe (LD-Nu) was developed to visualize the physical contact between LDs and nucleoli for the first time. Cell imaging results showed that LDs in both the cytoplasm and nucleus were susceptible to external stimuli, with cytoplasmic LDs being more affected. LD-Nu can serve as a powerful tool for further investigating the interaction mechanism between LDs and nucleoli in living cells.

Rational design of AIE-based carbazole derivatives for lipid droplet-specific imaging in living cells

Summary: In this study, a series of carbazole derivatives with D-A-pi-D structure were designed and synthesized, and their photophysical properties were investigated. Compound L7 showed the most favorable aggregation-induced emission behavior in ethanol/H2O mixtures and demonstrated high specificity and good biocompatibility in targeting lipid droplets, making it a promising candidate for specific visualization of lipid droplets at the cellular level.

Rational design AIE fluorescent probes for wash-free and lipid droplet specific imaging of fatty liver based on coumarin

Summary: This study developed a series of fluorescent probes based on the coumarin moiety, which exhibited aggregation-induced emission, ultrahigh signal-to-noise ratio, and wash-free properties. Compound B2 was successfully applied to real-time detection of dynamic lipid droplets in living cells. The imaging performance of compound B2 at the pathological tissue level in a mouse model with lipid droplet-related fatty liver was well investigated, indicating exceptional selectivity and resolution.

An environment-sensitive near-infrared (NIR) probe, TPA-DCPP NPs, was prepared to achieve multidimensional quantification of intracellular polarity, enabling accurate cancer cell discrimination and autophagy monitoring, offering ideas for the development of probes in complex biosystems.

Lysosome-Specific Coumarin-Based Fluorescent Bioprobes for in Vivo Polarity Sensing and Cancer Treatment

Summary: About 90% of cancer deaths are caused by the spread of cancer cells to distant organs. Lysosomes, due to their ability to induce cell death, have become attractive targets for cancer diagnosis and treatment. In this study, two fluorescent probes were constructed using coumarin derivatives. The results showed that these probes have a linear relationship with polarity change and can specifically identify and monitor tumor sites. They also showed a strong antiproliferation effect on cancer cells. These findings demonstrate the potential of these probes in the diagnosis and treatment of cancer.
