A Collection of RPA-Based Photoelectrochemical Assays for the Portable Detection of Multiple Pathogens

Summary: In this study, a photoelectrochemical (PEC) biosensor combined with recombinase polymerase amplification (RPA) technology (RPA-PEC) was proposed for rapid detection of multiple foodborne pathogens. The RPA was initiated on homemade three-dimensional screen-printed paper-based electrodes, forming a lab-on-paper platform. Using the DNA-PEC signaler, photocurrents were achieved after only 20 minutes of RPA, with superior detection performance compared to conventional agarose gel electrophoresis.

Copper: DNA extraction and solid phase detection agent for all-in-one molecular diagnostic device coupled with isothermal amplification

Summary: In this study, an all-in-one PMMA device integrating two novel techniques - DNA extraction employing a CuSO4/H2O2 system and DNA detection utilizing solid phase copper tape - coupled with LAMP is developed for on-site pathogen detection. The device uses the Fenton-like reaction to produce hydroxyl radicals that disrupt bacterial membranes and release DNA. The released DNA is amplified by LAMP and the corrosion of copper tape is used for DNA detection. The fabricated PMMA device integrates all the necessary functionalities for molecular diagnostics, and the equipment-free techniques coupled with the portable heater are promising for point-of-care testing in resource-limited environments.

Smartphone-based immunochemical sensor exploiting peroxidase-like activity of ligand-capped gold nanostars: A proof-of-concept detection of Mycobacterium bovis

The developed sensing mechanism showed promising results for the detection of Mycobacterium bovis, enabling detection at low concentrations.

Quercetin-Mediated Silver Nanoparticle Formation for the Colorimetric Detection of Infectious Pathogens Coupled with Loop-Mediated Isothermal Amplification

Summary: Here, a quercetin-mediated silver nanoparticle (AgNP) synthesis combined with loop-mediated isothermal amplification (LAMP) technique was used to detect SARS-CoV-2 and Enterococcus faecium using a foldable PMMA microdevice. The LAMP amplicons formed a complex with Ag+ ions, while the quercetin acted as a reducing agent and chelated with Ag+ ions, resulting in the formation of brown-colored AgNPs within 5 minutes. The method exhibited higher sensitivity than agarose gel electrophoresis and had a detection limit of 101 copies μL-1 for SARS-CoV-2 RNA and 101 CFU mL-1 for E. faecium.

Dual-mode visual detection strategies of viable pathogens for point-of-care testing

Summary: This study introduces a power-free foldable PMMA microdevice that integrates DNA extraction, amplification, and visual detection using colorimetric and aggregate formation modes. The microdevice consists of a reaction zone and a detection zone, connected by a sealing film for foldability. The presence of LAMP amplicons is detected by the oxidation of 4-AP, resulting in color change or aggregate formation. EthD-1 is introduced to identify viable bacteria and prevent further DNA amplification. Development of a POCT detection platform based on a locked nucleic acid-enhanced ARMS-RPA-GoldMag lateral flow assay

Summary: In this study, a novel genotyping point-of-care testing (POCT) rapid detection device was developed using a combination of locked nucleic acid (LNA)-amplification refractory mutation system (ARMS)-recombinase polymerase amplification (RPA)-GoldMag lateral flow assay (LFA) platform. The results of SNP typing detection and clinical validation showed that the LNA-ARMS-RPA-GoldMag lateral flow assay platform has high sensitivity, specificity, stability, accuracy, and feasibility. This new technology is expected to make a significant contribution to POCT rapid diagnosis and expand its application space, reflecting its clinical application value and development prospects.

Effects of the food colorant carmoisine on zebrafish embryos at a wide range of concentrations

Development of a Rapid Immuno-Based Screening Assay for the Detection of Adenovirus in Eye Infections
