TOP-LD: A tool to explore linkage disequilibrium with TOPMed whole-genome sequence data

Le Huang et al.

Summary: TOP-LD is an online tool that allows exploration of linkage disequilibrium (LD) using high-coverage whole-genome sequence (WGS) data from the TOPMed program. Compared to current LD tools, TOP-LD offers a more comprehensive representation of genetic variation, particularly for rare variants and specific populations. It includes a significantly larger number of variants and also includes information on structural variants. TOP-LD has the potential to facilitate a wide range of applications based on summary statistics and LD estimates.

Hedgehog Signaling: Linking Embryonic Lung Development and Asthmatic Airway Remodeling

Ling-Hui Zeng et al.

Summary: The development of the embryonic lung requires complex interactions, regulated by various signaling proteins. Hedgehog (Hh) signaling plays a vital role in lung development and its controlled regulation is crucial for maintaining healthy lung function. Aberrant Hh signaling is associated with several airway lung diseases, including bronchial or tissue fibrosis, lung cancer, and pulmonary arterial hypertension. The Hh signaling pathway is also linked to lung organogenesis and asthmatic airway remodeling.

Powerful, scalable and resource-efficient meta-analysis of rare variant associations in large whole genome sequencing studies

Xihao Li et al.

Summary: In this study, the researchers propose MetaSTAAR, a powerful and resource-efficient rare variant meta-analysis framework for large-scale WGS/WES studies. MetaSTAAR takes into account relatedness and population structure, and improves the power of rare variant tests by incorporating multiple variant functional annotations. The results show that MetaSTAAR performs well in large-scale rare variant meta-analysis and can be applied in biobank-scale cohorts.

Liability-scale heritability estimation for biobank studies of low-prevalence disease

Sven E. Ojavee et al.

Summary: Theory for liability-scale models of complex diseases provides a valuable tool for interpreting and comparing biological study results. This study proposes an extension of the existing liability-scale model theory suitable for estimating liability-scale heritability in biobank studies of low-prevalence diseases. By applying this new expression, the study demonstrates improved accuracy and stability in estimating low-prevalence disease outcomes, especially in cases of healthier sample populations.

Plasma Hsp90 levels in patients with systemic sclerosis and relation to lung and skin involvement: a cross-sectional and longitudinal study

Hana Storkanova et al.

Summary: According to the study, plasma levels of Hsp90 are increased in patients with systemic sclerosis compared to healthy controls, and are associated with increased inflammatory activity, worsened lung function, and extent of skin involvement in patients with diffuse cutaneous systemic sclerosis. In patients with systemic sclerosis-associated interstitial lung disease treated with cyclophosphamide, baseline Hsp90 levels can predict changes in DLCO over 12 months of therapy.
