The Elusive Structure of Levocetirizine Dihydrochloride Determined by Electron Diffraction

Summary: In this study, the crystal structure of Levocetirizine dihydrochloride was determined directly from crystalline powders extracted from commercially available tablets using 3D electron diffraction (3D ED)/MicroED technique. The results highlight the immense potential of 3D ED/MicroED for structure elucidation of components of microcrystalline mixtures, eliminating the need for growing large-size single crystals and using complementary analytical techniques, which is important for identification and primary structural characterization.

Atomic-level structure determination of amorphous molecular solids by NMR

Summary: In this study, the atomic-level ensemble structure of the amorphous form of the drug AZD4625 was determined by combining solid-state NMR experiments with molecular dynamics simulations and machine-learned chemical shifts. Preferred conformations and intermolecular interactions in the amorphous sample were analyzed to understand the stabilization of the amorphous form of the drug.

De Novo Crystal Structure Determination from Machine Learned Chemical Shifts

This study successfully determined the crystal structures of ampicillin, piroxicam, cocaine, and two polymorphs of the drug molecule AZD8329 using machine-learned isotropic chemical shifts generated on-the-fly to guide a Monte Carlo-based structure determination process.
