Cysteine synthase: multiple structures of a key enzyme in cysteine synthesis and a potential drug target for Chagas disease and leishmaniasis

Kate Sowerby et al.

Summary: Chagas disease and leishmaniasis are significant health problems caused by parasites. The study focuses on the cysteine biosynthesis pathway, which is essential for parasite survival. Crystal structures of cysteine synthase enzymes from Trypanosoma cruzi, Leishmania infantum, and T. theileri were determined, revealing the active site and potential binding sites for inhibitor design. These findings can aid in the development of new drugs and protein-protein inhibitors.

Use of benznidazole to treat chronic Chagas disease: An updated systematic review with a meta-analysis

Clara Crespillo-Andujar et al.

Summary: This study conducted a meta-analysis to evaluate the effectiveness of benznidazole in the treatment of Chagas disease. The results showed that benznidazole is recommended for children with Chagas disease based on serological response, but its efficacy in adults remains uncertain. Further clinical data is needed to assess the efficacy of benznidazole in both children and adults with Chagas disease.

Chagas Disease: Chronic Chagas Cardiomyopathy

Natalia Giraldo Echavarria et al.

Summary: Chagas disease is a tropical vector-borne infection caused by the parasite Trypanosoma cruzi, endemic in South and Central America but increasingly becoming a burden in the United States and Europe due to human migration. Most patients are asymptomatic during the acute phase, with around 30% progressing to detectable organ damage mainly affecting the cardiovascular and digestive systems in the chronic symptomatic phase, which can lead to Chagas cardiomyopathy, a leading cause of nonischemic cardiomyopathy in Latin America. Treatment during the parasitic infection is possible, but efficacy during the advanced chronic symptomatic phase remains uncertain. Further studies are needed to improve clinical outcomes and refine treatment recommendations.

Trypanothione Metabolism as Drug Target for Trypanosomatids

Maria Dolores Pineyro et al.

Summary: Chagas Disease, African sleeping sickness, and leishmaniasis are neglected diseases caused by pathogenic trypanosomatid parasites, which have significant impact in poor countries. Current treatment drugs are toxic, difficult to access, and may lead to drug resistance. Inhibiting essential parasite metabolic pathways unique from the host can lead to more efficient and safe drug development.

Biosynthesis, Quantification and Genetic Diseases of the Smallest Signaling Thiol Metabolite: Hydrogen Sulfide

Joanna Myszkowska et al.

Summary: Hydrogen sulfide (H2S) plays important roles in human health, with its disease-promoting effects depending on concentration and compartmentalization. There is substantial discrepancy in H2S concentrations determined by different techniques, with no definitive protocol for continuous, real-time measurement.

Transplantation for Chagas Heart Disease: a Comprehensive Review

Fernando Bacal et al.

Summary: Heart transplantation is now considered the best treatment for end-stage Chagas heart disease, with patients requiring a lesser degree of immunosuppression to balance reactivation risks. PCR-based techniques are being used more often to screen for Chagas disease reactivation in the first-year post-transplant. Multiple reports have demonstrated the feasibility and safety of heart transplantation for refractory Chagas cardiomyopathy.
