The novel sustained 3-hydroxybutyrate donor poly-D-3-hydroxybutyric acid prevents inflammatory bowel disease through upregulation of regulatory T-cells

Rimina Suzuki et al.

Summary: This study investigates whether poly-D-3-hydroxybutyric acid (PHB), a polyester of 3-hydroxybutyrate (3-HB), can be degraded by intestinal microbiota and serves as a slow-release agent of 3-HB to suppress the pathogenesis of inflammatory bowel disease (IBD). The results show that a PHB diet increases 3-HB concentrations in feces and blood, and suppresses weight loss and histological inflammation in an IBD model. Furthermore, PHB increases the accumulation of regulatory T cells in the rectum without affecting T cells in the spleen.

Theabrownin from Fu Brick Tea Improves Ulcerative Colitis by Shaping the Gut Microbiota and Modulating the Tryptophan Metabolism

Wuqi Yang et al.

Summary: Fu brick tea theabrownin (FBTB), produced by oxidative polymerization of tea polyphenols, can alleviate various diseases. This study explored the potential of FBTB in treating ulcerative colitis (UC) using a DSS-induced chronic UC mouse model. FBTB effectively reduced colonic damage and inflammation, improved colonic antioxidant capacity, and corrected the disrupted gut microbiota. FBTB elevated microbial tryptophan metabolites by promoting Lactobacillus growth and activated aromatic hydrocarbon receptors (AhRs) to repair the intestinal barrier. These findings demonstrate that FBTB alleviates UC by targeting the gut microbiota and the AhR pathway.

Huangshan Floral Mushroom Polysaccharide Ameliorates Dextran Sulfate Sodium-Induced Colitis in Mice by Modulating Th17/Treg Balance in a Gut Microbiota-Dependent Manner

Ming-yue Zou et al.

Summary: This study reveals the mechanism of how floral mushroom polysaccharide (FMPS) improves ulcerative colitis (UC) by investigating the coordinated interaction between intestinal microbes and the balance of intestinal helper T cell 17 (Th17) / regulatory T cell (Treg). The results show that FMPS can up-regulate the expression of certain proteins, reduce the secretion of inflammatory factors, regulate intestinal microorganisms, and improve the level of short-chain fatty acids (SCFAs). Importantly, FMPS-mediated improvement of UC depends on the restoration of Th17/Treg balance mediated by intestinal microorganisms.

Inflammatory Bowel Disease

Lia Pierson Bruner et al.

Summary: CD and UC, two forms of IBD, affect approximately 1% of the population and are characterized by chronic diarrhea, abdominal pain, and weight loss. The diagnosis relies on medical history, physical examination, laboratory tests, and endoscopic evaluation with biopsy. Advances in pharmacologic treatments have significantly improved outcomes, but chronic inflammation from IBD and immunosuppressive therapies raise the risk for certain conditions, necessitating collaboration between gastroenterologists and primary care providers for comprehensive patient care.

Emerging Roles of Gut Microbial Modulation of Bile Acid Composition in the Etiology of Cardiovascular Diseases

Tess Yntema et al.

Summary: Despite advancements in prevention and treatment, cardiovascular disease (CVD) remains the leading cause of death worldwide. Recent studies have highlighted the potential role of non-traditional factors, such as the gut microbiota and its metabolites, in CVD. Specifically, disturbances in the gut microbiota and altered bile acid metabolism have been associated with CVD development. Bile acids, apart from their role in lipid absorption and metabolism, have been found to act as signaling molecules and modulate cardiometabolic pathways, making them potential therapeutic targets in CVD.

Inflammation and malnutrition in inflammatory bowel disease

Sara Massironi et al.

Summary: Inflammatory bowel disease (IBD) has been on the rise worldwide, with impaired nutritional status in patients presenting in various forms such as malnutrition, sarcopenia, and micronutrient deficiency, as well as overweight, obesity, and sarcopenic obesity. Malnutrition can disrupt the gut microbiome, leading to an imbalanced state that triggers inflammation. Despite the clear link between IBD and malnutrition, little is known about the underlying mechanisms that promote inflammation and malnutrition. This Review explores the potential mechanisms behind the vicious cycle of malnutrition and inflammation, and their implications for clinical treatment.

Summary: Inflammatory bowel disease (IBD) is a chronic gastrointestinal disease with an imbalance in gut microbiota as a critical factor in its development. Gastrodia elata polysaccharides (GBP) have shown anti-inflammatory properties and potential for ameliorating IBD. In a study on DSS-induced IBD mice, GBP demonstrated the best therapeutic effect by reducing IBD symptoms and modulating the gut microbiota composition and metabolism. Investigation into the metabolic regulation of the gut microbiota provides a promising approach to evaluate the therapeutic effect of GBP on IBD.
