Spatial transcriptomics combined with single-cell RNA-sequencing unravels the complex inflammatory cell network in atopic dermatitis

Yasutaka Mitamura et al.

Summary: This study reveals the cellular crosstalk in the leukocyte-infiltrated area of AD skin through single-cell analysis and spatial transcriptomics. The results demonstrate unique distribution patterns and interactions among fibroblasts, dendritic cells, and macrophages in the skin lesions. Additionally, the expression levels of TNC and CCL18 are correlated with disease severity. These findings provide valuable insights for the development of improved treatments.

Single-cell profiles reveal distinctive immune response in atopic dermatitis in contrast to psoriasis

Bowen Zhang et al.

Summary: Single-cell analysis of atopic dermatitis and psoriasis patients revealed disease-specific T cell clusters and identified cell type-specific gene expression associated with inflammation signatures and disease severity. The study shed light on the complexity of inflammatory responses and provided valuable insights for further investigation and therapeutic approaches for these chronic inflammatory skin diseases.

ILC2s in skin disorders

Yasutomo Imai

Summary: This review discusses the tissue-specific properties of ILC2s in skin and their involvement in human skin diseases. In a steady state, cutaneous ILC2s contribute to tissue homeostasis. However, in patients with atopic dermatitis, IL-33 overexpression leads to activation of ILC2s and their production of type 2 cytokines.

Smad3 Promotes Cancer-Associated Fibroblasts Generation via Macrophage-Myofibroblast Transition

Philip Chiu-Tsun Tang et al.

Summary: This study identified a novel role of macrophage-myofibroblast transition (MMT) in cancer, showing that MMT cells can de novo generate cancer-promoting cancer-associated fibroblasts (CAFs). Through fate-mapping, RNA velocity, and pseudotime analysis, a subset of CAFs derived from macrophages was confirmed in the tumor microenvironment. Targeting Smad3, a key regulator in the MMT process, effectively blocked CAF formation and cancer progression in vivo, suggesting MMT as a potential therapeutic target for cancer immunotherapy.

Human β-defensin-3 attenuates atopic dermatitis-like inflammation through autophagy activation and the aryl hydrocarbon receptor signaling pathway

Ge Peng et al.

Summary: A study found that autophagy in keratinocytes is restrained in patients with atopic dermatitis (AD) and mouse models of AD. Human β-defensin-3 (hBD-3) alleviates the impairment of the tight junction barrier through activation of keratinocyte autophagy, and reduces skin inflammation. This suggests that autophagy contributes to the pathogenesis of AD, and hBD-3 could be used therapeutically.

Selective autophagy controls the stability of transcription factor IRF3 to balance type I interferon production and immune suppression

Yaoxing Wu et al.

Summary: IRF3, a critical transcription factor in antiviral innate immune signaling, is regulated by CALCOCO2 and PSMD14. CALCOCO2 promotes autophagic degradation of IRF3, while PSMD14 prevents such degradation, maintaining IRF3 activity and fine-tuning immune response against viral infection.

Developmental cell programs are co-opted in inflammatory skin disease

Gary Reynolds et al.

Summary: The skin establishes a complex cellular network early in embryonic development to provide biophysical and immunological protection. The study found an enrichment of innate immune cells in skin during the first trimester, as well as clonal expansion of disease-associated lymphocytes in atopic dermatitis and psoriasis. Additionally, a reemergence of prenatal vascular endothelial cell and macrophage cellular programs was observed in atopic dermatitis and psoriasis lesional skin.

IL-33-induced metabolic reprogramming controls the differentiation of alternatively activated macrophages and the resolution of inflammation

Maria Faas et al.

Summary: Uncoupling protein-2 and the transcription factor GATA3 were found to regulate the differentiation of AAMs in response to IL-33, leading to their polarization. IL-33 induced metabolic rewiring in macrophages, which subsequently triggered GATA3-mediated AAM polarization.

Susanne Mommert et al.

Summary: The study investigated the role of histamine in CCL18 production in human monocyte-derived M2 macrophages, finding that IL-10 upregulated H2R and increased CCL18 expression through H2R stimulation. This effect was similar in cells from atopic dermatitis patients compared to healthy controls.
