Recent developments in single electron/radical chemistry have allowed for site-specific editing of proteins. This review gives an overview of current methods that install such edits, focusing on radical C-C bond formation and its compatibility with proteins and cells, as well as the novel applications afforded by these methods. Despite technical challenges, radical C-C bond formation on proteins is a promising and rapidly growing field in chemical biology.

Pd(II)-Mediated C-H Activation for Cysteine Bioconjugation

Summary: Selective bioconjugation remains a significant challenge for synthetic chemists due to the high degree of functionality and stringent reaction conditions of biomolecules. This study reports the preparation of Pd(II) complexes for cysteine bioconjugation via a facile C-H activation process. These complexes demonstrate competitive bioconjugation efficiency and are synthesized using user-friendly methods and common Pd(II) sources, making them more cost-effective. Furthermore, these complexes do not require isolation and achieve high conversion efficiency and selectivity of a model peptide. They also show the ability to selectively arylate a single surface cysteine residue on a model protein substrate, further highlighting their utility.

Flavin Metallaphotoredox Catalysis: Synergistic Synthesis in Water

Antibody drug conjugate: the biological missile for targeted cancer therapy

The Chemistry Behind ADCs
