Synthesis of C-Aryl Glycosides by C-H Functionalization

Summary: In recent years, the synthesis of C-aryl glycosides through C-H functionalization has gained significant attention due to its efficiency and economy. This review focuses on the synthesis of C-aryl glycosides using C-H arylation of glycosides and C-H glycosylation of arenes, highlighting their regioselectivity and diastereoselectivity. It is observed that newly developed C-H glycosylation reactions exhibit better selectivity than traditional Friedel-Crafts glycosylation, with milder reaction conditions that are compatible with acid-sensitive protective groups. However, remote C-H glycosylation of aromatic hydrocarbons remains a relatively unexplored area that requires further research.

Benzo-Fused-Ring Toolbox Based on Palladium/Norbornene Cooperative Catalysis: Methodology Development and Applications in Natural Product Synthesis

Summary: This article summarizes the research progress of the author's team in the efficient assembly of common benzo-fused skeletons through Pd/NBE-promoted reactions. New annulation strategies using versatile bifunctional reagents were developed, including the use of epoxides and aziridines for alkylating reactions, olefinic alcohol-containing alkylating reagents for the formation of carbocycles, and cyclohexanone-containing amination reagents for the construction of N-containing bridged scaffolds. These new annulation reactions exhibited high selectivity, few steps, and atom economy, and have been applied in the total synthesis of medicines and complex natural products.

The resulting C8-aminated pyrrolo-phenanthridines exhibit strong fluorescence, and steric hindrance may prevent aggregation-caused quenching.

A Concise Total Synthesis of (-)-Berkelic Acid
