Revealing cell populations catching the early stages of human embryo development in naive pluripotent stem cell cultures

Summary: In this study, distinct cell populations coexisting with epiblast-like cells in 5iLAF naive human induced pluripotent stem cell (hiPSC) cultures were discovered and characterized. These cell populations closely resemble different cell types of the human embryo at early developmental stages. The presence of a totipotent eight-cell (8C)-stage-like cell population, as well as three populations analogous to trophectoderm cells at different maturation stages, and cells resembling primitive endoderm were detected. This provides an excellent opportunity to model the earliest events of human embryogenesis using 5iLAF naive hiPSC cultures.

Summary: FSHD is characterized by oxidative stress induced by DUX4, leading to metabolic dysfunction and impaired mitochondrial function. Increased mitochondrial ROS levels in FSHD muscle cells are associated with elevated steady-state mitochondrial membrane potential. DUX4 triggers mitochondrial membrane polarization, resulting in mitochondrial ROS generation and apoptosis.

Summary: Embryonic genome activation (EGA) is critical for embryonic development, but our understanding of the regulatory mechanisms of human EGA is still incomplete. In this study, a short-pulsed expression of DUX4 in primed hESCs activated an EGA-like gene expression program in up to 17% of the cells, while retaining cell viability. These DUX4-induced cells resembled eight-cell stage blastomeres and were named induced blastomere-like (iBM) cells.
