EVs-mediated delivery of CB2 receptor agonist for Alzheimer's disease therapy

Low Doses of beta-Caryophyllene Reduced Clinical and Paraclinical Parameters of an Autoimmune Animal Model of Multiple Sclerosis: Investigating the Role of CB2 Receptors in Inflammation by Lymphocytes and Microglial

Summary: This study investigated the protective effect of low doses of beta-Caryophyllene (BCP) on experimental autoimmune encephalomyelitis (EAE) mice, a chronic model of multiple sclerosis (MS). The results showed that BCP can reduce inflammation and improve pathological and clinical defects in EAE mice by modulating the immune system. BCP also increased the levels of anti-inflammatory cytokine IL-10 and decreased blood inflammatory cytokines. Therefore, low doses of BCP may be suggested as a therapeutic approach for MS with potent anti-inflammatory effects and possibly lower toxicity.

New Insights into Bitopic Orthosteric/Allosteric Ligands of Cannabinoid Receptor Type 2

Summary: We have developed a new generation of ligands (JR compounds) that target the CB2R. These compounds combine the pharmacophoric portion of CB2R positive allosteric modulator (EC21a) with that of CB2R selective orthosteric agonist (LV62). Among the tested compounds, JR22a showed dualsteric behavior as a CB2R ligand. Computational studies confirmed the binding mode of JR22a at CB2R, and its potential to prevent neuroinflammation was investigated using a human microglial cell model.

Summary: The design and synthesis of dualsteric/bitopic agents that can interact with both orthosteric and allosteric binding sites presents a novel approach in medicinal chemistry. In this study, we designed and characterized the first CB2R heterobivalent bitopic ligands, which displayed selective targeting of CB2R and functional selectivity for the cAMP signaling pathway. Additionally, the most promising bitopic ligand FD-22a showed anti-inflammatory and antinociceptive activity in in vitro and in vivo models, respectively. Computational studies further confirmed the bitopic nature of these compounds.
