Integrated multimodality microscope for accurate and efficient target-guided cryo-lamellae preparation

Summary: Cryo-electron tomography is a revolutionary technique for resolving the structure of subcellular organelles and macromolecular complexes in their cellular context. However, the sample preparation step hampers its application. In this study, cryo-CLIEM technique was developed, which integrates three-dimensional confocal imaging with focused ion beam-scanning electron microscopy for efficient preparation of lamellae containing target structures. This approach has been shown to be generally applicable and will contribute to more applications of cryo-electron tomography.

Characterization of mApple as a Red Fluorescent Protein for Cryogenic Single-Molecule Imaging with Turn-Off and Turn-On Active Control Mechanisms

Summary: Single superresolution microscopy provides a powerful tool for studying biological structures below the optical diffraction limit. Imaging at cryogenic temperatures reduces photobleaching and enables correlation with cryogenic electron tomography. However, a limitation for samples maintained at 77 K is the limited number of fluorophores known to undergo efficient turn-on and turn-off mechanisms. This study discovers a novel turn-off mechanism in mApple, a red emitting fluorescent protein, which enables localization of many individual molecules in initially bright samples, but the density of active emitters is limited by inefficient turn-on. Bulk excitation and emission spectroscopy reveals the complex nature and environmental sensitivity of states observed in red fluorescent proteins.

Correlating cryo-super resolution radial fluctuations and dual-axis cryo-scanning transmission electron tomography to bridge the light-electron resolution gap

Summary: Visualization of organelles and their interactions with other features in the native cell remains a challenge in modern biology. We introduced cryo-scanning transmission electron tomography (CSTET) which enables high-resolution 3D imaging on a micron scale, and demonstrated the utility of cryo-SRRF nanoscopy and deconvolution processing in enhancing precision and resolution. The integration of cryo-SRRF with deconvolved dual-axis CSTET provides a versatile workflow for studying unique objects in a cell.

Membrane protein isolation and structure determination in cell-derived membrane vesicles

Summary: Traditional methods for determining the structure of integral membrane proteins involve using detergents or polymers to extract them from cell membranes. However, in this study, we demonstrate a new approach of isolating and determining the structure of proteins in membrane vesicles derived directly from cells. Using this method, we were able to determine the structures of the ion channel Slo1 from both total cell membranes and cell plasma membranes at high resolutions. The results provide insights into the stability and interactions of the Slo1 channel in the plasma membrane environment, as well as uncovering previously unresolved regions and additional ion binding sites.
