Understanding nanomedicine treatment in an aggressive spontaneous brain cancer model at the stage of early blood brain barrier disruption

Summary: Personalised nanomedicine is a developing field that shows promise in targeting aggressive cancer with minimal side effects. In this study, researchers used magnetic resonance imaging techniques to determine the optimal stage at which to administer nanomedicine for treating brain tumors. The results showed that nanoparticle-delivered doxorubicin effectively reduced tumor growth and increased tumor apoptosis. This study highlights the potential of polyethylene-based nanoparticles in treating tumors with disrupted blood-brain barrier.

Landscape of Epidermal Growth Factor Receptor Heterodimers in Brain Metastases

Summary: This study investigated the relationship between brain metastases and drug treatment in HER2+ breast cancer patients. The results showed that brain metastases from breast, ovarian, lung, and kidney cancers have higher levels of HER2-HER3 dimers compared to other primary tumor types. However, traditional HER2-targeted therapies, such as pertuzumab, have limited inhibitory effects on HER2-HER3 dimerization in the presence of excess neuregulin, which may explain the occurrence of brain metastases.

Development and Evaluation of Competitive Inhibitors of Trastuzumab-HER2 Binding to Bypass the Binding-Site Barrier

Summary: Our group has developed a strategy to increase antibody penetration in solid tumors by transiently inhibiting antibody-antigen binding. The strategy was validated experimentally and showed increased efficacy of antibody-based therapeutics in HER2+ xenograft bearing mice. We also developed a mechanistic pharmacokinetic model to predict within-tumor exposure of the antibody and a pharmacodynamic component to predict the therapeutic effect. The study has important implications for optimizing antibody-cytotoxin penetration and efficacy in solid tumors.

Modeling Brain Metastasis by Internal Carotid Artery Injection of Cancer Cells

Summary: Brain metastasis is a significant cause of morbidity and mortality in cancer patients, and animal models play a critical role in studying therapeutic interventions. Existing brain tumor xenografting methods lack consistency, and intracardiac injection can result in unintended extracranial growth. This study presents a mouse model of brain metastasis generated by internal carotid artery injection, which produces consistent intracranial tumors.

This study explores a drug predosing strategy to suppress the mononuclear phagocyte system and improve the pharmacokinetics of a model nanoparticle system in mice.
