Summary: Cancer is a deadly disease characterized by abnormal cell growth and invasion. Current treatment options lack selectivity and have side effects, driving the search for better anticancer drugs. Copper-containing compounds have shown excellent activity against cancer cells. The redox activity of copper ions, combined with bioactive ligands, contributes to their cytotoxicity. Copper can induce DNA damage and reactive oxygen species production, while light-assisted excitation of copper complexes generates reactive species for photodynamic therapy. In vivo and in vitro studies have demonstrated the effectiveness of copper ions against various cancer cell lines.

Summary: A new copper (II) complex with neocuproine and triazine has been synthesized and characterized. The complex was characterized using UV spectra, FT-IR, EPR, and thermal analyses. The binding mechanism of the Cu (II) complex with CT-DNA was determined using UV spectra, fluorescence spectra, and cyclic voltammetry techniques. In vitro cytotoxicity assays showed that the Cu (II) complex exhibited potent cytotoxic behavior.

This article provides an overview of anthracycline use in cancer therapy, discussing their mechanisms of action, toxicity, and chemoresistance, as well as approaches to improve their effectiveness, reduce toxicity, and overcome resistance.

Drug-Tolerant Persister Cells in Cancer Therapy Resistance

Summary: One of the challenges in cancer treatment is the development of acquired resistance to therapy, which is responsible for a large percentage of cancer-related deaths. This review focuses on persister cells, a subpopulation of drug-tolerant cells in tumors, and discusses approaches to target them. The review highlights the characteristics of a specific persister cell subpopulation with high levels of aldehyde dehydrogenase (ALDH) activity and their role in drug resistance and cancer progression. The interactions between ALDH-high persister cells and the tumor microenvironment are also explored. Therapeutic modulation of this subpopulation is proposed as a strategy for more effective cancer management.

Summary: In this study, the interaction between 21 Casiopeinas and DNA was investigated using gel electrophoresis, UV-vis spectroscopy, and circular dichroism spectroscopy. Results showed that Casiopeinas can interact with DNA through intercalation or in the minor groove, and the mode of interaction is influenced by the substituents on the diiamine ligands.

Copper induces cell death by targeting lipoylated TCA cycle proteins

Summary: Copper is an essential cofactor for all organisms, but excessive copper can lead to cell death. We found that copper-dependent cell death mechanism is different from known mechanisms, and it is related to mitochondrial respiration. Copper directly binds to lipoylated components of the tricarboxylic acid cycle, causing protein aggregation and loss of iron-sulfur cluster proteins, ultimately leading to cell death.
