Hyperelastic characterization reveals proteoglycans drive the nanoscale strain-stiffening response in hyaline cartilage

Summary: Degenerative diseases like osteoarthritis lead to cartilage matrix deterioration, affecting tissue function. Atomic force microscopy (AFM) is commonly used for measuring mechanical properties, but linear elastic contact models ignore the nonlinear nature of soft materials, impacting clinical studies. This study compared classical Hertzian theory with five hyperelastic models to measure cartilage ECM and found that the Fung model demonstrated the best fit. By selectively degrading ECM components, the study also revealed that proteoglycans drive the nonlinear response of cartilage ECM. These findings contribute to the biomechanics community's understanding of two-parameter material models for micro-indentation of soft biomaterials.

Comparison of the effects of AgNPs on the morphological and mechanical characteristics of cancerous cells

Summary: In this study, the morphological and mechanical effects of silver nanoparticles (AgNPs) on A549 and SMMC-7721 cancer cells were investigated using atomic force microscopy (AFM). The AFM measurement, including force-volume mode and force spectroscopy mode, was utilized to characterize the morphological and mechanical properties of the cancer cells influenced by AgNPs. The results showed significant variations in the mechanical properties, such as height, roughness, adhesion force, and Young's moduli, of the two cancer cell types with different concentrations of AgNPs. This research provides insights into the classification and characterization of the effects of AgNPs on cancer cells for disease therapy using AFM methodologies.

Jamming Transitions in Astrocytes and Glioblastoma Are Induced by Cell Density and Tension

Summary: Collective behavior of cells is crucial for wound healing, embryonic and tumor development, and can undergo a transition from a migratory unjammed state to a static jammed state depending on cell density and interactions. This study investigated the collective migration dynamics of astrocytes and glioblastoma cells and explored cell adhesion, traction, and mechanics using various techniques. Results showed that astrocytes remained in a non-migratory jammed state, while glioblastoma cells exhibited migratory unjammed behavior. Switching between jammed and unjammed states could be induced by altering the balance between cell-cell adhesion and tension via inhibition of ROCK or myosin II. These findings are important for understanding glioblastoma cell infiltration and developing novel anti-migratory drugs and treatment strategies.

Experimental extraction of Young's modulus of MCF-7 tissue using atomic force microscopy and the spherical contact models

Nanomechanical Signatures in Glioma Cells Depend on CD44 Distribution in IDH1 Wild-Type but Not in IDH1R132H Mutant Early-Passage Cultures

Summary: Atomic force microscopy (AFM) has recently become a valuable tool in biomedicine, allowing for the analysis of cancer cells and their microenvironment to better understand tumor invasion and progression. In this study, high-resolution AFM mapping was used to analyze the nanomechanical properties of glioma cell cultures with different IDH1 R132H mutation statuses. The results showed that IDH1 R132H mutant cells exhibited increased stiffness and elasticity modulus compared to IDH1 wild-type cells. Additionally, CD44+/IDH1wt cells were found to be more rigid and stiffer than CD44-/IDH1wt cells. However, there were no significant nanomechanical differences between CD44+/IDH1 R132H and CD44-/IDH1 R132H cells. These findings suggest that quantitative nanomechanical mapping holds promise for the accurate analysis of cell populations and personalized treatment of glioma.

We observed that the perinuclear region of cancer cells was softer relative to the lamelopodial stiffness, and a significant stiffening phenomenon was found in the thin lamellipodial region.

Analysis of phellinus igniarius effects on gastric cancer cells by atomic force microscopy

Summary: This study used an atomic force microscope to quantitatively detect the effects of Phellinus linteus on gastric cancer cells. It was found that Phellinus linteus can change the morphology of cells, increase their height and surface roughness, reduce cell adhesion force, and increase the elasticity modulus. However, it has less impact on gastric epithelial cells. These results demonstrate that Phellinus linteus can be used as a preferred anticancer drug for the treatment of gastric cancer cells.

Application of self-organizing maps to AFM-based viscoelastic characterization of breast cancer cell mechanics

We propose the use of self-organizing maps (SOMs) as unsupervised artificial neural networks to analyze mechanical measurements on breast cancer cells treated with different substances that affect estrogen receptor signaling.

Atomic force microscopy-based assessment of multimechanical cellular properties for classification of graded bladder cancer cells and cancer early diagnosis using machine learning analysis

Machine learning was employed to train a cancerization-grade classifier for classification of BC cells at different grades. Compared with traditional diagnostic methods, the proposed non-invasive prognosis has higher accuracy with fewer cellular properties as inputs. It provides a potential means for early detection of cancerization with high sensitivity and specificity.

Characterization of Breast Cancer Aggressiveness by Cell Mechanics

Summary: In healthy tissues, cells are mechanically stable. However, during cancer progression, cancer cells undergo changes in their mechanical properties, becoming softer and more fluid-like. This is associated with cytoskeletal remodeling, altered adhesion properties, increased cell proliferation, and enhanced cell motility. In our study, we examined the mechanical properties of different breast cancer cells using atomic force microscopy force spectroscopy. Our results revealed that the mechanical properties of the cells are linked to their malignancy, with tamoxifen-resistant MCF-7 cells being the softest and MCF10A cells being the stiffest. Confocal fluorescence microscopy further demonstrated differences in actin network organization and focal adhesion localization. These findings contribute to our understanding of the complex alterations involved in cancer development.

Summary: High-speed atomic force microscopes (HS-AFMs) are capable of visualizing dynamic phenomena at nanoscale resolution with high temporal resolution. However, the complexity and cost of HS-AFMs, coupled with the lack of published information on open-source HS-AFM controllers, have hindered their widespread adoption in academia and industry. Furthermore, the limited imaging area of HS-AFMs restricts their potential applications. This study introduces an open-source controller that enables a low-cost simplified HS-AFM system with significantly improved tip-sample velocity and imaging area. The proposed system successfully assesses the severity of atopic dermatitis by measuring the nanotexture of human skin corneocytes. By providing affordable access to high-throughput nanoscale imaging, the low-cost HS-AFM system based on the open-source controller enhances the research capabilities of resource-limited institutes and contributes to the expansion of the HS-AFM research community.
